| Objectives:Chaiqinlidan Compound Granule (CQLD) is a new Chinese traditional medicine which achieved by modern pharmaceutical technology mainly used to take care of cholecystitis. This thesis studied pharmacological action of CQLD and its mechanism through a series of experiments to provide theoretical evidence for clinical trials.Methods:In this thesis, pharmacodynamics and possible mechanisms of CQLD were studied in treating cholecystitis. A lot of classical models were established to evaluate the anti-inflammatory effect and it's probable mechanism of CQLD like model of mouse ear edema, model of rat paw edema induced by carrageenin, Acetic acid has been injected into the mice belly to increase the permeability of capillary vascular and to induce mice leucocyte migration. The analgesic action of CQLD was studied by two standerd models:acetic acid induced writhing and hot-plate test with mice be used as tempals. Choleretic effect of CQLD has been observed by testing the flow rate of gall in rats. Then a pyogenic cholecystitis model has been made by injecting E.coli into rabbits'gall bladder after their bile ducts were ligated partially. After 7-day feeding, the concentration of ALT, AST, and direct bilirubin in plasma, the concentration of total cholesterol, total bilirubin and direct bilirubin in bile and the stress of sphincter of Oddi have been tested. Therapeutical action of CQLD on the injuring cholecysts was also observed.Results:1.The anti-inflammatory experiments showed that:CQLD suppressed the mice ear edema induced by xylene and rats'paw edema induced by carrageenin obviously; It also obviously inhibited the secretion of IL-1, TNF-α, PGE2 and MDA in rats'paws. We also found that CQLD can also inhibit the capillary vascular permeability and suppressed the migration of leucocyte.2. The analgesic experiments showed that CQLD can reduce the number of writhing response due to pain caused by acetic acid in mice and remarkably extend the incubation period of licking foot caused by hot plate and enhance pain threshold.3. The choleretic experiments showed that CQLD can increase the flow rate of bile in rats in 90 min. It can obviously promote the secretion of bile in rats. 4. The therapeutic experiments of CQLD on pyogenic cholecystitis showed that CQLD lowered the concentration of ALT, AST, and direct bilirubin in plasma, declined the concentration of total cholesterol, total bilirubin and direct bilirubin in bile and relaxed the sphincter of Oddi. Pathological observation indicated that invasion of inflammatory cells was serious in sick rabbits. Immunohistochemical experiment showed that the concentration of TNF-αin rabbits'gall bladder have grown up, but there are no remarkable differences compared with normal rabbits.Conclusions:1. CQLD has excellent anti-inflammatory, analgesic and choleretic effects.2. CQLD can improve pyogenic cholecystitis in rabbits remarkably.3. It is probably that the anti-inflammatory effect of CQLD relates to its action to decrease the concentration of IL-1, TNF-α, PGE2 and MDA in inflammatory issue. |
