Expression Of MMP-2 And TIMP-1 In Rats Endometriosis Treated With Genistein

Objective:To explore the influence of genistein on the expressions of matrix metalloproteinase-2(MMP-2) and tissue inhibitor of metalloproteinase-1 (TIMP-1) in rats endometriosis(EMs).Methods:Sixty-eight Wistar rat in which endometriotic implants were induced were randomly divided into five groups, Group I (normal control group),Groupâ…¡(model group), Groupâ…¢(lower-dose Genistein), Groupâ…£(middle-dose Genistein) and Groupâ…¤(high-dose Genistein), All rats of Group3-5 continued to recive treatmemt for 12 weeks, and then were killed to assess the changes of implants size and for pathoological analysis HE stain was used to observe pathological changes of implants. The expressions of MMP-2 and TIMP-1 in the grafts were examined by immunohistochemical S-ABC method.Results:1. The implants'volume was no significantly difference (P> 0.05) in the groupâ…¢and groupâ…£; but there was significantly decreased in groupâ…¤(P<0.01);2. Pathology changes:Compared with the groupâ…¡, the epithelial layer of ectopic endometrium was less and the thickness of epithelial layer was thinner in trayment groups, and there was more effects in groupâ…¤;3. Campared with the groupâ… ,the expression rate of MMP-2 was significantly increased in groupâ…¡(P<0.05);Campared with the groupâ…¡, the expression rate of MMP-2 was no significantly differences in groupâ…¢and groupâ…£(P>0.05); but there was significantly decreased in group V than the those groupâ…¡(P<0.05);4. Campared with the groupâ… . the expression rate of TIMP-1 was significantly decreased in groupâ…¡(P<0.05);Compared with the groupâ…¡, the expression rate of TIMP-1 was no significantly differences in groupâ…¢and groupâ…£(P>0.05); but there was significantly increased in groupâ…¤than the those groupâ…¡(P<0.01).Conclusion:1.MMP-2 and TIMP-1 obviously up-regulate and down-regulate in ectopic endometrium and they may play an important role in the development of endometriosis;2.Genistein has a good therapeutic effect on rats'EMs. The mechanism may related with the genistein down-regulate MMP-2 expression and up-regulate TIMP-1 expression.
