| Objective:To observe the spatial learning and memory abilities and hippocampual plasticity of the temporal lobe epilepsy rats induced by lithium-pilocarpine;To investigate the effects of ginsenoside Rd and Dan hong injection on the behavior and pathologic changes of the rats after the drugs'interventions and possible treatment mechanisms.Methods:Our research includes three parts:(1) In this study, the chronic spontaneous temporal lobe epilepsy rats models were established with lithium and pilocarpine intraperitoneally.24 temporal lobe epilepsy rats were randomly divided into 2 groups:TLE group (NS 10ml/kg, intraperitoneal injection),and Ginsenoside Rd intervention group (Ginsenoside Rd 2mg/kg,intraperitoneal injection).in addition,6 rats were assigned to the normal control group.Morris water maze test was conducted for valuating the learning and memory abilities in each groups. The changes of mossy fiber sprouting in hippocampus were examined by Timms staining. (2)30 temporal lobe epilepsy rats were randomly divided into 2 groups:TLE group (NS 10ml/kg, intraperitoneal injection), and Ginsenoside Rd intervention group (Ginsenoside Rd 2mg/kg,intraperitoneal injection).In addition,15 rats were assigned to the normal control group.Morris water maze test was conducted for valuating the learning and memory abilities in each groups. The changes of 5-HT in hippocampus were examined by immunohistochemistry staining. (3) The temporal lobe epilepsy rats models were established with lithium and pilocarpine intraperitoneally.50 temporal lobe epilepsy rats were randomly divided into 2 groups:TLE group (NS 10ml/kg, intraperitoneal injection), and Dan hong injection intervention group (Dan hong injection 10ml/kg,intraperitoneal injection).In addition,25 rats were assigned to the normal control group. Each group of rats were randomly divided into five different time points:Id,3d,7d,15d and 30d.Nissel staining was conducted for valuating the survival neurons in hippocampus CA1 in each groups. The changes of GFAP were examined by immunohistochemistry staining.Results:(1) Compared with normal control group,rats in TLE group showed worse learning and memory abilities(P<0.05)and larger number of mossy fiber sprouting in dentate gyrus of hippocampus (P<0.05);Compred with TLE group Ginsenoside Rd intervention group had significantly improved on the spatial learning and memory abilities (P< 0.05) and showed smaller number of mossy fiber sprouting in dentate gyrus of hippocampus (P<0.05). (2) Compared with normal control group,rats in TLE group showed worse learning and memory abilities(P<0.05)and fewer expression of 5-HT in hippocampus CA1 (P<0.05); Compred with TLE group Ginsenoside Rd intervention group had significantly improved on the learning and memory abilities (P<0.05)and showed more expression of 5-HT in hippocampus CA1 (P<0.05). (3) Compared with normal control group,rats in TLE group showed fewer survival neurons (P<0.05)and more expression of GFAP in hippocampus CA1 (P<0.05); Compred with TLE group,Dan hong injection intervention group had significantly improved the quantity of survival neurons (P<0.05)and showed fewer expression of GFAP in hippocampus CA1 (P<0.05).Conclusion:(1) Ginsenoside Rd may play an important role in alleviating the injury on the spatial learning and memory abilities in TLE by inhibiting the mossy fiber sprouting in hippocampus. Ginsenoside Rd probably can interfere with the onset and development of the chronic spontaneous temporal lobe epilepsy. (2) Ginsenoside Rd plays an important role in alleviating the injury on the learning and memory abilities in TLE by increasing the expression of 5-HT in hippocampus CA1. (3) Dan Hong injection can help to relieve the extent and block the formation and development of seizures in rats with chronic temporal lobe epilepsy by alleviating the neuronal loss and reducing the expression of GFAP in hippocampus CA1. |
