ObjectiveTo explore the inhibitory effect of disodium cantharidinate and Vitamin B6 combined with radiotherapy against hepatocellular carcinoma cell line HepG2.MethodsThe inhibitory effect of HepG2 treated with disodium cantharidinate and Vitamin B6 injiction was measured by MTT assay.radiosensitivity was evaluated By using a clonogenic assay.The early stage apoptosis was detected with Annexin-V-FITC/PI double labeled assay,and the apoptosis was obversed by fluorescence microscope。Resultsdisodium cantharidinate and Vitamin B6 group, radiotherapy treatment group and disodium cantharidinate and Vitamin B6 combined radiotherapy group can inhibited the proliferation of HepG2.Compared with the control group, disodium cantharidinate and Vitamin B6 can inhibited the proliferation of HepG2 significantly, Inhibition of disodium cantharidinate and Vitamin B6 combined radiotherapy group on HepG2 was significantly higher than disodium cantharidinate and Vitamin B6 group or radiotherapy treatment group.suggest that there is a synergistic effect between disodium cantharidinate and Vitamin B6 and radiotherapy on HepG2. Clone formation assay suggested that disodium cantharidinate and Vitamin B6 enhenced sensibility of HepG2 to radiotherapy. Conclusiondisodium cantharidinate and Vitamin B6 can inhibit greatly proliferation, induce apoptosis,increase radiosensitivity of human hepatocellular carcinoma cells. So it is valuable in clinic. |
