Objective To investigate the effection of disodium cantharidinate to hepatic carcinoma cell HepG2 . We detect the inhibition of cell,proliferation,expression of survivin,the activity of caspase-3 and the change of cell cycle. Through these we can analyse the influence of disodium cantharidinate to HepG2 and its molecular mechanism.Methods HepG2 cells were treated with different concentration of disodium cantharidinate. The rate of cell inhibition,expression of survivin,cell cycle profile and activity of Caspase-3 were respectively detected by MTT assay,immunocytochemistry,flow cytometry and chromatometry.Results Disodium cantharidinate could inhibit the cell reproduction of HepG2 . 72 hours after treatment, the rate of cell inhibition was respectively 39.28%,56.62%,81.75%,91.27%. 48 hours after intervention, the cells at G2/M phase of HepG2 were increased and the extent of arrest had a positive correlation with drug concentration. HepG2 had the positive expression of survivin , disodium cantharidinate could inhibit its expression and active the activity of caspase-3.Conclusion Disodium cantharidinate can inhibit the cell reproduction of HepG2 and induce the G2/M arrest. The expression of survivin is down regulated by disodium cantharidinate and the activity of caspase-3 is actived at the same time. Disodium cantharidinate may through its arresion of G2/M,down-regulation of survivin and Caspase-3 dependent manner to influence the multiplication of HepG2. |
