| ObjectiveRenal interstitial fibrosis is a common passage to the final renal failure, which has become one of the hot topic in the renal disease research. Bone marrow-derived mesenchymal stem cells(MSCs)has the abilities to deferentiate to all kinds of renal cells so as to participate in the renal repair or renew after damage.MSCs has been utilized to some common animal model of renal interstitial fibrosis, but we can not search any utilization of MSCs to this kind of animal model.In this study, bone marrow-derived MSCs of Wistar rats were separated and cultured in vitro, and then transfected with Ad-GFP. Renal renal interstitial fibrosis was established and Ad-GFP transfected MSCs were injected into rats with renal interstitial fibrosis. Finally, we observe whether MSCs can immigrate and localize in the rat's kidney, and what effect to the renal function and pathology changes of the kidney.Materials and methodsWe separated and cultured MSCs of Wistar rats in vitro and identified its CD34 and CD90 with immunhistochemistry; we transfected MSCs with Ad-GFP and examined the infective rate with cytometry; We transfected MSCs with the most suitable MOI and transplanted them to the rats.We selected 25 3 months years'old Wistar rats and randomly divided them into 2 gruopsto established the animal model; Then we used the successful model to be transplanted MSC; Finally, we observed the rats for 1 months before we got the urine and kept the kidney for HE, PAS, Masson and immunhistochemistry and etc. Results1.We separated MSCs from the rats and cultured them for 7-9 days before we passaged the cells. Immunhistochemistry demonstrated that the CD34 was negertive and CD90 was positive.2. When MOI=400, the transfect efficiency is 54.1%3.We gived 100mg/kg/d adnine to rats to successfully established the renal interstitial fibrosis animal model.4.The immunfluresene showed that many green positive cells in the renal interstial area and the yellow positive rate is 31.4%.5.Cr and BUN was higher in transplanted group than the onotransplanted group and the 24h urin protein is higher, so as to the pathology score. a-SMA show stronger than the nontransplanted group.ConclusionIn the Adnine-induced renal interstitial fibrosis rats model, the Ad-GFP transfected MSCs through tail vein can get to the kidney and differenate to epithelial cells of renal tubules but they can not obstruct the process of renal interstitial fibrosis. It can even execrates the lesion of renal fibrosis and renal gramarula and make the renal function worse. |
