| Objective:To establish a rat model similar to the metabolic abnormalities of human type 2 diabetes mellitus, plant Titanium implants,then observe and measure the morphologic parameters of around the implant. Furthermore, by the experiment out of the body, find the factors which is closely related to the heal of the DM Patients'bone issue, pursue the possibility mechanism which is influenced of the osseointegration of implants.Method:1. Establish DM rat model:divide the 44 six-month old male rats into 4 Groups randomly(DM implanting group, normal implanting group, DM control group, blank control group). DM implanting group and DM control group are induced to DM model using the Streptozotocin (STZ). After 4 weeks, DM implanting group and the normal implantin group plant the implants into the Epiphyseal end of the right tibia near, while the remaining two groups do the sham-operation.2. Observation Histology and Imageology:After 8 weeks, kill the rats by drawing blood of inferior vena cava, measure the changes of the AGEs by fluorospectrophotometer, observe the form, BMD and the state of the osseointegration of bones around implants and analyse the relationship between DM and the formation of the bone issue around the implants and the osseointegration of implants.3. The observation of Immunohistology:analyse the relationship of the BG level, the AGEs level and the expression of RAGE in the bone issue around the implanted places.Results:1. the heal condition of implants:DM implanting group and normal implanting group are all not loosened, the redness of soft tissue and fester under periost is appeared in 3 examples of DM group, normal group do not have these situations.2. the result of X-ray image:tibia cortex around the implanted places is thin which is partly absorbed, the width of ossis is increased, the texture of spongy bone is sparse and becoming thinner in the DM implanting group and the DM control group. Tibia cortex and bone density of the last two groups is regular in The implanted bodies of the normal group in which ossis is narrower compared to the first two groups. the texture of spongy bone density is compact and even. The peri-implant density of the DM implanting is lower. 3. The change of the BMD, the AGEs, the BG and the weight. there is statistically significant in the four groups(P<0.05), besides, there is statistically significant each of two groups (P<0.05).4. Tibia RAGE immunohistochemistry all have positive expression in 4 groups. The expression of the DM implanting is the stronget, even some is sliced shape. Normal implanting group also have RAGE positive expression, but it is divergence and light. The result of positive cell ratio and positive cell average optical in the image shows that the value in DM implanting group is obviously higher than normal implanting group and DM control group is higher than bland control group. The difference has statistical significance (P<0.05). Furthermore, DM implanted group is higher than DM control group. The difference also has statistically significant (P<0.05).Conclusion:The result of this research shows that serum AGEs ofâ…¡type DM rats is obviously increased and tibia cortex of bone is thin which is partly absorbed, the width of ossis is increased, the texture of spongy bone is sparse and becoming thinner. The expression of bone issue RAGE potential aroundâ…¡type rats implanted places is strong and extensive and the colouration is deep and obviously higher than control group.
