ObjectiveTo identify the existence of orexin receptor 1 (OX1R) in obese rat pancreas, clarify the effect and mechanism of OX1R on body weight and metabolism index in obese rats.METHODSEstablish obese rat model induced by high-fat diet, serum glucose, triglyeride and insulin were respectively measured by Mini-Blood Glucose meter, Biochemical enzymatic method and chemiluminescent immunoassay. The expression of OX1R in pancreas was identified by Immunohistochemistry. After intracerebroventricular infusion of orexin receptor 1 antisense oligodeoxynucleotide (OX1R-PS-ODNs), detect the effect on OX1R in rat pancreatic tissue with obesity.RESULTSAfter feeding 8 weeks, body weight, serum glucose, insulin, triglyeride and cholesterol of high-diet rats were higher than control group (p<0.05).The expression of OX1R located in cytoplasm of pancreas Islet B-cells.The expression of OX1R in obese rat pancreas decreased about 16.4% compare with normal group (p<0.05).After obese rats were treated by OX1R-PS-ODNs for 72 hours,the expression of OX1R in pancreas reduced about 34.1% compare with normal group (p<0.05)CONCLUSIONSOX1R expressed in pancreas Islet B-cells. Hypothalamic orexin system can intervene the expression of 0X1R in rat pancreatic tissue with Alimentary obesity, and participate the occurrence of obesity and the metabolic regulation of serum glucose, blood fat and insulin. |
