Objective:Application of tissue microarray and immunohisto-chemical methods, PTEN, and Ezrin (Ades protein) in non-small cell lung cancer were detected, analyzed with clinicopathologic factors and the relationship between survival time of patients, to investigate the both the occurrence and development in the role of NSCLC, for the clinical treatment of NSCLC and prognosis of new theories.Methods:Produced tissue microarray by 80 cases of non-small cell lung cancer (NSCLC),with 20 cases of inflammatory pseudotumor as a control group.Followed by immunohistochemistry (MaxvisionTM) detected the expression of PTEN and Ezrin protein in tissue microarray and control group. SPSS16.0 statistical software for data analysis,use X2 test, Fisher cut indeed probability, Spearman rank correlation analysis the relationship between PTEN, Ezrin protein expression and clinicopathological features of NSCLC. Use Kaplan-Meier method in univariate survival analysis, survival curves were compared using Log-rank test; PTEN, Ezrin expression and other possible factors affecting the prognosis of NSCLC patients were analyzed by COX proportional hazards regression model. Results:1. Organization of the chip can be seen neatly organized lattice, rendering satisfaction.80 organizations, including four points lattice completely off-chip, organizations lattice falling rate of 5%(4/80), part of the organization there curly and some missing points.2. PTEN, Ezrin protein expression rate in NSCLC tissues were 59.2%,81.6%in inflammatory pseudotumor of tissue were 85.0%,55.0%, PTEN, Ezrin in NSCLC and inflammatory pseudotumor of tissues,There was significant difference (p<0.05).3. PTEN expression in NSCLC and sex, age, pathological type, histological grade, tumor size and other factors not related (p> 0.05). In the TNM staging for the Phaseâ… +â…¡,â…¢+â…£of PTEN expression in tumor tissue in patients with positive rates were 71.8%, 45.9%; in the absence of lymph nodes and lymph nodes associated with the tumor tissue of patients with positive rates of PTEN expression 69.8%,45.5%; compare the differences were statistically significant (p<0.05).4. Ezrin expression in NSCLC and different gender, age, pathological type, histological grade, tumor size and TNM stage was not significant (p> 0.05). Without lymph node metastasis and lymph node metastasis in patients with Ezrin expression were 69.8%,97.0%, the difference was statistically significant (p<0.05).5. NSCLC tissue PTEN and Ezrin protein expression was not correlated (r=0.089 p= 0.444).6. Univariate analysis showed that, TNM stage, lymph node metastasis, PTEN expression, Ezrin expression is associated with the prognosis of patients with lymph node metastasis and TNM stage later, PTEN negative expression and Ezrin expression indicates a poor prognosis.7. COX multivariate survival analysis:TNM stage, PTEN and Ezrin expression is an independent prognostic risk factors in NSCLC patients (p<0.05).Conclusion:1. PTEN in NSCLC tissues was significantly lower than inflammatory pseudotumor, Ezrin expression in NSCLC tissues was significantly higher than inflammatory pseudotumor, may be used as markers of clinical diagnosis of NSCLC.2. The expression of PTEN and Ezrin in NSCLC relate whether there is lymph node metastasis in NSCLC patients. Suggest that PTEN and Ezrin proteins may be involved in NSCLC development, invasion and metastasis process.3. PTEN and Ezrin expression is one of the factors affecting prognosis of patients with NSCLC. Detection of PTEN and Ezrin protein expression in judging the prognosis NSCLC might have important value.4. PTEN and Ezrin protein expression in NSCLC patient have no correlation.
