| As a serious disease hazard to human life and health, malignant tumor has naturally become the focus of human society. This paper is focused on the research in diagnosis and treatment of two common malignant tumor by constructing multi-functional nano materials.1,Up-converting phosphor immunochromatography test for quantitative detection of AFPUp-converting phosphors (UCP) were synthesized by using oleic acid as a stabilizing agent through a facile hydrothermal method. Based on the surface modification and activation, it can be connected with bioactive molecules. With integration of immunochromatography and UCP marked technology, a rapid and simple method of AFP quantitative detection is established. We evaluated the sensitivity, specificity, accuracy and precision of this method, the concordance rate between UCP and Axsym was analyzed through 100 clinical sera. It was found that typical analysis time was less than 20 minutes. And the sensitivity of the test was 5 ng/ml. No cross reaction was found when the method was used to detect other tumor markers (including CEA, CA199, FER, AFU and NSE). Furthermore, accuracy and precision of the assay can meet the needs in clinical treatment. Compared to the method of Axsym by determination coefficient (R2=0.9908), it shows good correlation. 2,Synthesis of Multifunctional siRNA complexes and the interference study of K562 cellsThe silica-coated fluorescent nanoparticles(SiO2/FITC) were prepared via Stober method. And the characteristics of these nanoparticles were analysised with TEM, UV and fluorescence spectrophotometer. Then polyethyleneimine(PEI) was modified to the surface of SiO2/FITC to obtain multifunctional siRNA complexes. The result showed that the synthesized SiO2/FITC nanoparticles were monodispersed, with a good fluorescence intensity and photostability. And the complexes can be successfully transferred into K562 cells through endocytosis, which was constructed by coupling SiO2/FITC with siRNA. It can effectively decrease the level of bcr/abl mRNA. In this paper, SiO2/FITC not only work as a good vector but also exhibit photostability for cell marking, which can be used to monitor the delivery and transfection of the siRNA.
