Metabolic syndrome is a cluster of interacting risk factors of cardiovascular disease and type 2 diabetic mellitus,which comprises abdominal obesity,blood lipid disorders,inflammation,insulin resistance or full-blown diabetes.The cause of MS remains obscure,but it's wildly accepted that both genetic and environmental factors contribute to it, and the high-fat high-calory diet and induced activity play an important role as environmental factors.C57BL/6J mice have a high susceptibility to diet-induced obesity,type 2 diabetes,and atherosclerosis.So we choose C57BL/6J as a genetic background mouse strain and high-fat high-calory purified diet as inducing factor to make up MS animal model.Nowadays the drug treatments of MS focus on the concrete disorders,without integrant effective strategies.So we also evaluated the effects of three group traditional Chinese compound prescription so called natural combinatorial chemical libraries which characterized of multitarget.We found,the mice in high-fat high-calory diet(model group) show significantly higher body weight(BW) and demonstrated central obesity(P<0.001),compared with mice in chow diet(control group) after 1 week on respective diet fed.After 4 weeks,mice in model group began to show hyperglycemia(P<0.01) and the curve of intraperitoneal glucose tolerance testing(IPGTT) illustrated impaired glucose tolerance.After 6 weeks,the model group appeared hyperinsulinemia,revealing the state of insulin resistant.The model group showed higher serum TG,HDL-C,TC,LDL-C and FFA after 10 weeks(P<0.001).We also found the BW of model mice having positive significant correlation with above index of disordered glucose and lipid metabolism. All of these demonstrated we established mouse model of MS successfully. After the mice model had been established,we evaluated the therapeutic effect of three traditional Chinese compound prescriptions.Then we found the erchen decoction could degrade the weights of body liver and visceral fat,lower the levels of FBG,120min blood glucose in IPGTT test,and FFA TG LDL-C in lipid metabolic disorders' profile significantly,Zhihuang decoction had similar effectives,but the mice depilated.It may result from dystrophy as the component Dahuang has the effect of diarrhea and purging heat.Pingweisan decoction could only lower BW FBG and TG slightly.Briefly, by just feeding C57BL/6J mice on high fat diet for 10 weeks,we successfully estabolished MS mouse model,which easy to reproduce,short period,stable and mimicking human MS progression well.And basing on the model,we screened erchen decoction as an effective treatment for the whole metabolic disordes. This revealed it might be a potent treatment to invigorate spleen to remove dampness and promote qi and activate blood circulation.
