| ObjectiveSHUANGHUA Granules (SHG) is independently developed with Xiushan honeysuckle as the main raw material.We have studied the effect and mechanism of SHG against liver injuries, in order to observe the protection function and the function mechanism of SHG fighting against the acute liver injured mice induced respectively by carbon tetrachloride and alcohol,and furnish the theoretical basis for developing and exploiting new drug from SHG against liver injuries.Methods1. The effect of SHG on acute liver injured mice induced by carbon tetrachlorideThe experimental, male,60 mice cleaned by ICR,were randomly divided into 6 groups by weight:normal group, model group, drug positive group(Tiopronin,250mg·Kg-1·bw-1), SHG group low,middle, high groups(1.25,2.5,5.0g.Kg-1·bw-1).According to the dose, Intragastrically infused with SHG and Tiopronin twice everyday, normal and model group Intragastrically infused with normal saline. After 14 days latter, All groups except normal of acute hepatic injury were set up by giving intraperitoneal injection of 0.1%CCl4 (0.2ml/10g).And 18 hours later all mice were put to death. Blood was taken from the back eye pit, and then centrifugation with 3000r/min by 15 Minutes to separate the serum, measured the activities of AST and ALT in the serum. Measured liver index.A soma piece of liver was taken 0.5cm far from the edge of lobes of liver for making frozen sections and observation the changes of morphology. Another hepatic tissue was made to be 10% liver homogenate and measured the SOD activity and MDA content.2. The effect of SHG on acute liver injured mice induced by alcoholThe experimental, male,60 mice cleaned by ICR,were randomly divided into 6 groups by weight:normal group, model group, drug positive group (Bifendate, 150mg·Kg-1·bw-1), SHG group low,middle, high groups(1.25,2.5,5.0g.Kg-1·bw-1) According to the dose, intragastrically infused with SHG/Bifendatatum twice everyday, normal and model group intragastrically infused with normal saline. All groups except normal of acute hepatic injury were set up by giving intragastrically infused of 56%alcohol (16ml-1·bw-1) at 11:00 everyday. After 14th day work finished, draw the materials and detection the index by the method 1.Results1. The effect of SHG on acute liver injured mice induced by carbon tetrachlorideResults showed that compared the model control group with normal control group, liver index increased significantly, activities of serum ALT and AST increased significantly, the contents of MDA in the liver tissue were significantly increased, and SOD activity was markedly decreased (P<0.01).The model control group liver tissue is turgid the centrol the liver sub-phyllode showed spread and poly-incomplete necrosis and fat denaturation.T inflammation cells have been infiltrated. The mice acute hepatic injury experimental model was built very successfully.Compared the groups, who had low,middle,high groups(1.25,2.5,5.0 g·Kg-1·bw-1)with model control group, we can see the liver index was decreased(P<0.05). The serum ALT and AST decreased significant (P<0.05). SOD obviously increased, and had obvious difference compared to the model group (P<0.01), and MDA in liver decreased in SHG high, middle, low group, and had obvious difference compared with the model group (P< 0.05).HE section showed that:the drug- treated groups relieved the necrosis,swelling,steatosis, inflammatory of hepatocytes. The effects of SHG (5.0g·Kg-1·bw-1) was better than other treatment groups, and there were no obvious hepatocytes necrosis and steatosis in the group.2. The effect of SHG on acute liver injured mice induced by alcoholResults showed that compared the model control group with normal control group, liver index increased significantly, activities of serum ALT and AST increased significantly, the contents of MDA in the liver tissue were significantly increased, and SOD activity was markedly decreased (P<0.01).HE section showed that in model group liver cell cord disordered, there were patches necrosis of liver cell with inflammatory collections around central vein and area of focal necrosis, hepatielobule's structure was unclear, cell swollen, and there were many lipid droplet vacuoles. The mice acute hepatic injury experimental model was built successfully.Compared the groups, who had low,middle,high groups(1.25,2.5,5.0 g·Kg-1·bw-1),with model control group, we can see the liver index was decreased(P<0.05). The serum ALT decreased significant (P<0.05). and the activities of AST obvious difference compared with the model group (P<0.05), SOD obviously increased, and had obvious difference compared to the model group (P<0.05), and MDA in liver decreased in SHG high,middle,low group, and had obvious difference compared with the model group(P< 0.05,).HE section showed that:the drug- treated groups relieved the necrosis of hepatocytes and inflammatory infiltration. SHG could inhibit fatty degeneration and reduced the number of fat droplet. The effects of SHG (5.0 g·Kg-1·bw-1) was better than other treatment groups, and there were no obvious hepatocytes necrosis and steatosis in the group.ConclusionSHG can resist acute liver injury in mice induced by CCl4.SHG have protective effects for acute liver injured mice induced by alcohol. |
