Expressions And Significances Of P14ARF,E2F1,CXCR7,NDRG1 In The Human Colorectal Tumor | | Posted on:2011-02-27 | Degree:Master | Type:Thesis | | Country:China | Candidate:Q Shen | Full Text:PDF | | GTID:2144360302494242 | Subject:Pathology and pathophysiology | | Abstract/Summary: | PDF Full Text Request | | Objective:To investigate the expression of P14ARF,E2F1,CXCR7,NDRG1 in human colorectal tumor,to clear out the influences of the genes on the tumorigenesis and development of colorectal cancer.Methods:P14ARF,E2F1,CXCR7 and NDRG1 were detected at their protein levels by immunohistochemical(IHC) staining and image analysis technique (IAT) in colorectal normal mucosae,colorectal adenomas,colorectal cancers with no regional lymph nodes metastasis,colorectal cancers with regional lymph nodes metastasis and their corresponding regional lymph nodes metastatic sites.Results:1.The positive expressions of P14ARF decreased gradually from colorectal normal mucosa tissues,to adenoma tissues,to cancer tissues;these expression differences among 3 classes were statistically significant either(P<0.05);The expression differences between cancers with lymphonode metastasis and cancers without metastasis was not notable (P>0.05);There was no obvious differences of the expression of P14ARF protein with clinicopathologic characters(P>0.05).2.The positive expressions of E2F1 decreased gradually from colorectal normal mucosa tissues,to adenoma tissues,to cancer tissues;these expression differences among 3 classes were statistically significant either(P<0.05);The expression differences between cancers with lymphonode metastasis and cancers without metastasis was not notable (P>0.05);There was no obvious differences of the expression of E2F1 protein with clinicopathologic characters(P>0.05).3.The expressions of CXCR7 protein was gradually increased from normal colorectal mucosa tissues,to colorectal adenoma tissues,to colorectal cancer tissues with no regional lymph nodes metastasis,but it was not statistically significant(P>0.05);And the expressions of CXCR7 protein between colorectal cancer tissues with regional lymph nodes metastasis and normal colorectal mucosa tissues/colorectal adenoma tissues/colorectal cancer tissues with no regional lymph nodes metastasis,and had significantly difference(P<0.05);The expressions of CXCR7 were not correlated with some clinicopathological indices(P>0.05) except tumor TNM staging.4.NDRG1 proteins were gradually increased from colorectal normal mucosa tissues,to colorectal adenoma tissues,to colorectal cancer tissues with no regional lymph nodes metastasis,to colorectal cancer tissues with regional lymph nodes metastasis;There was significantly difference of the expressions of NDRG1;The expressions of NDRG1 were not correlated with some clinicopathological indices(P>0.05) except tumor TNM staging.5.It was positive correlation between the expressions of P14ARF and E2F1, of CXCR7 and NDRG1(rs=0.278 & rs=0.334);It was passive correlation between the expressions of P14ARF and NDRG1,of E2F1 and NDRG1(r_s=-0.373 &r_s=-0.238).Conclusions:The proteins P14ARF and E2F1 have some contribution to suppressing colorectal carcinogenesis,and the protein dysfunction was just one early action of colorectal carcinogenesis.The protein CXCR7 had some contribution to promote metastasis of colorectal cancer;and had correlations with tumor progression;and was no relations with the early action of colorectal tumorigenesis;NDRG1 may play an important positive role in colorectal carcinogenesis and progression.There was positive correlation between the expression of P14ARF and E2F1 in colorectal carcinogenesis.There was positive correlation between the expression of CXCR7 and NDRG1 during invasion and metastasis of colorectal cancer. | | Keywords/Search Tags: | P14ARF, E2F1, CXCR7, NDRG1, Colorectal neoplasms, Image Analysis, Immunohistochemistry | PDF Full Text Request | Related items |
