The Expression Of CGMP Dependent Protein Kinase â…¡ (PKGâ…¡) In Human Normal Digestive Tract' Tissue And Corresponding Malignant Tumors' Tissue

Purpose:Detection of cGMP dependent protein kinaseⅡ(cGMP dependent Protein KinaseⅡ,PKGⅡ) in human normal digestive tract'tissue and corresponding malignant tumors'tissue,and explore PKGⅡin cancer development role and clinical significance.Methods:(1) Immunohistochemical SP method was applied to detect PKGⅡexpression in 110 cases of human normal digestive tract'tissue and 110 cases of corresponding malignant tumors'tissue.(2) Reverse transcription polymerase chain reaction(RT-PCR) was applied to assay PKGⅡexpression in 50 cases of human normal gastric tissue and 50 cases of primary gastric cancer tissue;RT-PCR was applied to assay PKGⅡexpression in 30 cases of normal esophageal tissue and 30 cases of primary esophageal cancer tissue.(3) To analyze the relationship between the clinical data and experimental results.Results:(1) immunohistochemistry results:①In 110 cases of digestive tract' cancer tissue,PKGⅡnegative expression rate was 72.73%(80/110), while PKGⅡnegative expression rate in 110 cases of digestive tract' normal tissue was 10.00%(11/110).②In 50 cases of gastric cancer tissue, PKGⅡnegative expression rate was 80.00%(40/50),while PKGⅡnegative expression rate in 50 cases of normal gastric tissue was 6.00% (3/50);In 30 cases of esophageal cancer tissue,PKGⅡnegative expression rate was 56.67%(17/30),while PKGⅡnegative expression rate in 30 cases of normal esophageal tissue was 3.33%(1/30);In 18 cases of colon cancer tissue,PKGⅡnegative expression was 72.22% (13/18),while PKGⅡnegative expression rate in 18 cases of normal colon tissue was 22.22%(4/18);In 12 cases of rectal cancer tissue,PKGⅡnegative expression rate was 83.33%(10/12),while PKGⅡnegative expression rate in 12 cases of normal rectal tissue was 25.00%(3/12).③In 10 cases of highly differentiated gastric cancer tissue,negative expression rate of PKGⅡwas 40.00%(4/10),in 16 cases of moderately differentiated gastric cancer tissue,negative expression rate of PKGⅡwas 81.25%(13/16),in 24 cases of poorly differentiated gastric cancer tissue,negative expression rate of PKGⅡwas 95.83%(23/24),PKGⅡnegative expression rate was significantly different(P<0.05) among the groups of gastric cancers tissue with different degrees of differentiation; In 11 cases of highly differentiated esophageal cancer tissue,negative expression rate of PKGⅡwas 27.27%(3/11),in 9 cases of moderately differentiation esophageal cancer tissue,negative expression rate of PKGⅡwas 66.67%(6/9),in 10 cases of poorly differentiated esophageal cancer tissue,negative expression rate of PKGⅡwas 80.00%(8/10). PKGⅡnegative expression rate was significantly different(P<0.05) among the groups of esophageal cancer tissue with different degrees of differentiation.④In 21 cases of early gastric cancer tissue,negative expression rate of PKGⅡwas 71.43%(15/21),in 29 cases of advanced gastric cancer tissue,PKGⅡnegative expression rate was 86.21% (25/29),between different clinical stages of gastric cancers,PKGⅡnegative expression rate was no significant difference(P>0.05).Of these, in 16 cases of early esophageal cancer tissue,PKGⅡnegative expression rate was 50.00%(8/16),in 14 cases of advanced esophageal cancer tissue, PKGⅡnegative expression rate was 64.29%(9/14),between the different clinical stages of gastric cancers,PKGⅡnegative expression rate was not significantly different(P>0.05).(2) reverse transcription polymerase chain reaction(RT-PCR) test results:①In normal gastric tissues'group,PKGII/GAPDH((?)+S) was 1.532±0.285,while in primary gastric cancer tissue group PKGII/GAPDH((?)+S) was 0.456±0.239;.Two guoups were significantly difference(P<0.01).②In normal esophageal tissue'group, PKGII/GAPDH((?)+S) was 1.841±0.314,primary esophageal cancer tissue'group,PKGII/GAPDH((?)±S) was 0.392±0.212.Both groups were significantly difference(P<0.01).Conclusion:Primary digestive tract'malignant tumors,PKGⅡexpression rate was lower than normal digestive tract'tissue.Primary gastric cancer,the expression of PKGⅡwas associated with tumor'degree of differentiation. The lower degree of differeentiation,the expression of PKGⅡis lower. But its expression with age,sex,clinical stages and lymph node metastasis has no matter.Primary esophageal cancer,the expression of PKGⅡwas associated with tumor'degree of differentiation,the lower degree of differentiation,the expression of PKGⅡis lower;primary esophageal cancer,the expression of PKGⅡwa associated with lymph node metastasis,lymph node metastasis,PKGⅡexpression is low,but its expression with age,sex,clinical stages has no matter.PKGⅡreduction or lack in primary digestive tract'cancer may be an important factor in the development of malignant tumors,is a sign of aggravation in gastric cancer and esophageal cancer.
