BACKGROUND AND OBJECTIVE : Gastric cancer is one of the most common malignant tumors in the world, accounting for the second rank in the carcinomatous decease. The annual mean death rate of gastric cancer is 16 per one hundred thousand in our country, which accounts for the first rank among the gastrointestinal tumors. The total five-year survival rate of gastric cancer is about 10%. At present, the diagnosis of carcinoma is mainly based on endoscopic histopathological examination. However, this process is restricted by many factors. Therefore, looking for a convenient, fast and accurate method for early diagnosis of cancer is significant. The tissue and cell always become carcinoma from molecule. Raman spectroscopy which can reflect the subtle structure of molecules, is able to detect the early changes of gastric cancer that won't show any clinical signs and now it has become a potential way of the early diagnosis of gastric cancer. This study is aimed to explore the difference of serum Raman spectrum between gastric cancer patients and healthy controls, the difference of preoperative and postoperative serum Raman spectrum in gastric cancer patients, and the difference of Raman spectrum between gastric cancer cell culture medium and 1640 culture medium ,so as to provide scientific proof for the clinical use of Raman spectrum and demonstrate that Raman spectroscopy could be a new method for the screening of gastric cancer, the monitoring of prognosis and the judgment of therapeutic effctiveness.METHODS: The specimens as follows were collected: the serum preoperation,post-operation for three days of gastric cancer patients , nomal controls, the culture medium of SGC7901 gastric cancer cells culture for 24,48 and 72 hours and 1640 culture medium.RESULTS: At wavelength of 585, 633, 656, 674, 707, 772, 776 , 798nm, the Raman peak of gastric cancer patients was higher than that of the normal. P<0.01, the difference is significant. Study of the Raman spectrum of gastric cancer patients preoperation and post-operation for three days shows that the Raman peak of post-operation group was obviously lower than the Raman peak of pre-operation group. The numerical value of the p was 0.003, 0.010, 0.020, 0.017, 0.034, 0.036, 0.032, 0.016 and the statistical difference is significant. From Raman spectroscopic study of the culture medium , the Raman peak repeatability at 585, 633, 656, 674, 707, 772, 776 and 798nm could be found .But the Raman peaks at 633, 656, 674nm can only be found in the culture medium of gastric cancer cells. At the 8 wavelengths mentionded above, the Raman peak was positively correlated with the culture time .CONCLUSION:â‘ The Raman peak of gastric cancer patients was higher than that of the normal.â‘¡The Raman peak of post-operation group was obviously lower than the Raman peak of pre-operation group.â‘¢The differences of the spectrums between the culture medium of gastric cancer cells and 1640 medium are significant. The Raman spectrum of culture medium of gastric cancer cells rises as time goes by.â‘£The technique of Raman spectrum can reflect the differences of the spectrums. It is considered effctive for the screening of gastric cancer, the monitoring of prognosis and the judgment of therapeutic effect.
