Depression is a mood disorder or affective disorders that make person feel sad or hopeless for an extended period of time which is caused by a variety of reasons. Depression is a common psychiatric disease, its incidence and prevalence rate in the gradual growth. The complexity of mechanism in depression is generally believed that related to Neurotransmitters, neuro-endocrin, Etc. At present, the academic community affirmed the participation of 5-hydroxytryptamine and acetyl cholinesterase is an important part of its pathogenesis. a large number of 5-hydroxytryptamine and acetyl cholinesterase exist in Hippocampus where is the vital structures and is recognized with learning, memory, emotions, endocrine and other closely related areas.Depression belong to the scope of diseases such as epilepsy, Melancholia in traditional Chinese medicine, it was been recaorded by"Huangdi's Orthodox Classic","Danxi's Mastery of Medicine". In the Ancient literatures, about 130 acupuncture points have been using in treatment of mental illness, such as Siguan acupoints, 13 ghost acupoints. Acupuncture (including Electro-acupuncture) in the treatment of depression obtain a reliable effect after long-term observations since the 50's the 20th century, and has aroused wide attention both at local and abroad. Acupuncture is a green therapy without any side effects. Compared to acupuncture and traditional antidepressants, there are a lot of advantages, such as simple, cheap and easily accepted by patients. Therefore, the application of this research is very high value.ObjectiveTo investigate the influence of Electro-acupuncture on 5-serotonin(5-HT) and Acetyl cholinesterase (AchE)'s changes in hippocampus of the rat model with chronic stressed-induced depression, try to find out the depression mechanisms for the treatment of Electro-acupuncture(Ea).MethodsChoose 35 healthy Wistar rats and divide them into Normal control(Nc) group(10) and Establishment of model(Em) group(25)randomly. Depression models are established by lonely raising and chronic unpredictable mild stress for twenty-one days, then divide Em group into 2 groups named Ea treatment (11)and model group(11)randomly after the success of Establishment of model. The former are given by Baihui, Sanyinjiao acupoints(2Hz,1mA) 20 minutes 1 time per day for 14 days and then made immunohistochemistry and measure the integrated option density(IOD)of 5-HT and AchE in hippocampus of rats, using image analysis system for analysis.ResultsCompared with normal control group, results of Open-field test and Volume of sucrose-intake in Establishment of model group decreased(P<0.001). IOD on 5-HT and AchE: the comparison of model group and normal control group, model group decreased(P<0.05). the comparison of model group and Ea group, model group decreased too(P<0.05); the comparison of Ea and normal control group had no differences.1. Open-field test resultsCrossing-movement(Cm) times:Compared to Nc group, Em group's Cm times of Open-field test decreased(P<0.001)at the 21st y of modeling time, and the difference had statistical significance.Compared with two different times,including the 7th day and before the modeling time ,14th day and before the modeling time, 21st day and before the modeling time, Em group's Cm times decreased(P<0.01), the differences has statistical significant. And the longer model times,the fewer Cm times. Nc group's rats had no significant change at different time points and no statistical significance.Rearing-movement(Rm)times:Compared to Nc group, Em group's Rm times of Open-field test decreased(P<0.001)at the 21st y of modeling time, and the difference had statistical significance.Compared with two different time points, including the 7th day and before the modeling time ,14th day and before the modeling time, 21st day and before the modeling time, Em group's Rm times decreased(P<0.01), the differences has statistical significant. And the longer model times, the fewer Rm times. Nc group's rats had no significant change at different time points and no statistical significance.2. Volume of sucrose-intake testCompared to Nc group, Em group's volume of sucrose-intake test decreased(P<0.001)at the 21st y of modeling time, and the difference had statistical significance.3. Comparison of IOD of 5-HT and AchE in hippocampus among 3 groups5-HT:The overall compared among Nc group, Ea group and model group(F=8.00,p=0.0017), the differences had statistical significance. Using SNK method at alpha = 0.05 level, compared with model group and Nc group, compared with model group and Ea group,the differences both have statistical significance, compared with Ea group and Nc group, the difference has no statistical significance.AchE:The overall compared among Nc group, Ea group and model group, (F=3.68,p=0.00375),the differences had statistical significance. Using SNK method at alpha = 0.05 level, compared with model group and Nc group, compared with model group and Ea group,the differences both have statistical significance, compared with Ea group and Nc group, the difference has no statistical significance.Conclusion1. Cm times of Open- field test reflects the activity of animals,Rm times of open-field test reflects the behavior and curiosity to explore of animals. Through the chronic stressed-induced depression 21days then analysis of the open field test data before and after the model time, we come to the conclusion: Em group rats are changes in open-field test, compared to Nc group, their behaviors, activities, and curiosities to explore are reduced obviously.2. Rats prefer sucrose water to drink under normal circumstances. Compared with Nc group and Em group's volume of sucrose-intake in two time points, it can be seen:Em group animals's volume of sucrose-intake were reduced. Their preferences on the sucrose water is far less than the Nc group without any stimulating.3. Depression cause the 5-HT and AchE reduced in hippocampus, Electro-acupuncture treatment can be achieved therapeutic purposes by repairing of damaged neurons, stabling the structure, improving the 5-HT, AchE level in the hippocampus of model rats.
