Establish rat gastrocnemius denervation and reinnervation model; using constructed FIV lentivirus vector transduct AMF gene into primary cultured rat myoblast, then the cell culture supernatant was collected for AMF purification; purified AMF solution was regularly injected into gastrocnemius of neurotization model. Nerve conduction velocity (NCV) of tibial nerve; (peak to peak value)PPV,area under the curve (AUC) of CMAP were measured 8w, 16w, 24w after injection finished, the end plate in gastrocnemius were counted for evaluation of mucle function. Meanwhile, another group, in which non-transfected myoblasts used,was set to be control. Through out the experiments, in the AMF group, a significant improvement in muscle functional restoration can be observed; and more end plates can be count. No trend of tumorigenesis was found in each tissue examed.The results showed that after AMF injection, denerved muscle function was more likely to recover both in velocity and quality. The endplates in exprimental group were also taking a quantitative advantage in restoration. Most of the evidence suggest that AMF behaved positively in neuromuscular reparation, while the mechanism has not been fully revealed and confirmed. It was a potential protein to be study further for clinical therapies.
