| Polysaccharide is a kind of nature biomacromolecule. The main poly- saccharides of Phaeophyta are polymerides consisted of aldose and ketose. Polysaccharides are essential constituents of all living organisms and play pivotal roles in cell identification, differentiation and proliferation both in physiological and pathological conditions. Seaweed polysaccharide is the total name of various kinds carbohydrate biomacromolecule in the marine algae. The evident properties of the commixture are water-solubility and stickiness. Seaweed polysaccharide commixture includes Sodium Alginate, Laminarin, and Polysaccharide. Many researchers have shown that nature polysaccharide possesses biological activity of immunomodulatory and anti- cancer. Consumed mainly as sea food, Undaria Pinnatifida is a species of the economically important brown algae and has been traditionally cultivated in China, and there is abundant production in Dalian every year, so we select Undaria Pinnatifida as our subject.Apoptosis, or programmed cell death (PCD), is an evolutionarily con- served, genetically controlled process and a homeostasis process responded to superfluous or dysfunctional cells. The proteins involved in apoptosis are related with the mechanism of tumor. We extracted and purified the Undaria Pinnatifida polysaccharides (UPPS); optimizd the conditions of the extrac- tion course. And we analyzed the compound of the UPPS further. We demon- strated the antitumor effects of UPPS in lung cancer A549 cells. The results showed: 1) The Undaria Pinnatifida polysaccharides (UPPS) extraction parameters were according to the optimized conditions: fifty grammas of Undaria Pinnatifida dried powder were steeped in 1500 ml of distilled water for 5 h at 90℃. Trypsin and Sevage reagent were used to remove the remnant proteins ; 2) UPPS were further purified by DEAE Cellulose-52 and Sephadex G-100 chromatography to afford three polysaccharide compo- nents; 3) We demonstrated the antitumor effects in lung cancer A549 cells by MTS assays, flow cytometry of tumor cells. The results showed that UPPS significantly inhibited the proliferation of A549 cells and induced the apoptosis in a dose-dependent manner.4) UPPS-treated A549 cells exhibited a marked decrease in Bcl-2 and Survivin expression which play the key roles in apoptosis through the immuocytochemistry . |
