Expression Of Interleukin-17 And IL-17R In Spleen Tissues Of Septic Rats And The Effect Induced By Ulinastatin Injection

Objective To explore the expression of IL-17,IL-17R in spleen tissue of septic rats and the effect induced by ulinastatin injection.Method 1.The animal model of sepsis rats was replicated the cecal ligation and puncture(CLP) animal model.Fivty male SD rats were randomly divided into 4 groups:the blank control group,the sham-operated group,the sepsis group and the ulinastatin treatment group(the sepsis group followed by injecting ulinastatin(30000ku/kg.d).2.After 72hours,the livability of rats were counted and blood samples were cramped out though abdominal aorta for CRP analysis;the expression of IL-17 in spleen tissue were tested by immunohistochemistry in four groups;the pathologic construction changes of Spleen tissues were determined by HE staining;WESTERN BLOT was used to test the expression of IL-17R protein.Results 1.the survival rate in the sepsis group and the ulinastatin treatment group is 33.3%and 56.3%,whereas there were no death in the blank control group and the sham-operated group;from the pathological section of HE staining,we can see severe inflammatory reaction in the sepsis group,the inflammatory reaction could be prominently improved by ulinastatin,no abnormality be seen in the the blank control group and the sham-operated group.2.there was a similar positive rate of IL-17,IL-17R and similar levels of CRP in the the blank control group and the sham-operated group in rats spleen tissue,obviously,there was no signifecance(P＞0.05) Meanwhile,the levels of CRP and positive rate of IL-17,IL-17R were hihger in the sepsis group compared with the the ulinastatin treatment group.Also,,there was significance between the sepsis group,he the ulinastatin treatment group,in the the blank control group and the sham-operated group.3.there was significant correlation between IL-17 and IL-17R.Conclusion 1.there was a higher expression of IL-17 and IL-17R.in the sepsic rats; 2.the secretion of IL-17 and expression of IL-17R protein in the spleen tissues of sepsis rats may be eased by Ulinastatin injection,the same as the inflammatory reaction.
