Objective: To explore the mechanism of the treatment of Menopausal syndrome with Catgut Embedding in Points with the study on the effect of the expression of ERamRNA in the female Model of Menopausal Syndrome rats' Pituitary.Methods:The female SD rats with ovaries removed, were adopted as the animal model of menopausal pathology, and were divided randomly into ovariectomized model group (A,n=9), catgut embedding in points group (B,n=11),drug group (C,n=10).At the same time, built sham model group (D,n=9) and normal group (E,n=8).Group B was treated with catgut embedding in points, once every 5 days, four times in all. Acupoints chosen in group B were as follows: Guanyuan (CV4), bilateral Sanyinjiao (SP6) and bilateral Shenshu (BL23). Group C was infused with Nilestriol 5ml/kg (1mg/kg) once every 5 days, four times in all. Group A,D and E were infused with water 5ml/kg once every 5 days, four times in all. After being treated 4 times in succession, all of the experimental animals were put to death on the 5th day after the last treatment, and collected serums and samples, The use of The use of real time polymerase chain reaction (RT-PCR) determination of the rats in each group expressed in different pituitary ERamRNA; anatomy from pituitary, hypothalamus, uterus, single, unilateral adrenal gland, and precision that check the above calculation of organ wet weight of organ weight coefficient.Result: (1) model group and the embedding group, drug group, sham-operated group, the normal expression of ERamRNA comparison group were significantly lower, the difference was significant (P <0.05), and one of the drug group, sham-operated group, normal group very significant differences (P <0.01); compared with the embedding group: normal group significantly increased the expression of ERamRNA, there is significant difference (P <0.05), drug group, sham-operated group, between the normal group were not significantly difference (P> 0.05). (2) pituitary weight coefficients: comparison between the groups were not significantly different (P> 0.05); the weight coefficient of the hypothalamus: the model group and the embedding group, sham operation group, the weight coefficient of the hypothalamus were significantly reduced, the differences are there is significant (P <0.05), embedding group and drug group, the weight coefficient of the hypothalamus increased, the difference was not significant (P> 0.05); adrenal weight factor: drug group and model group, the adrenal weight factor significantly or high significant difference (P <0.05); uterine weight factor: the model group and the embedding group, drug group, sham-operated group, compared to normal uterus weight were significantly lower coefficient, the difference was significant (P <0.001 ), embedding the drug group compared with no significant difference (P> 0.05), embedding group and normal group difference was significant (P <0.05), sham-operated group compared with the normal group no significant difference (P> 0.05), while the drug group and normal group no significant difference (P> 0.05).Conclusion: (1) in rats after castration may be the cause of pituitary expression ERamRNA; Catgut Implantation can affect the neuroendocrine regulation of pituitary ERamRNA expression, the implementation of the regulation of pituitary function. (2) in rats after castration may cause the uterus, the hypothalamus gland and other organs of the aging, degradation, reducing the organ coefficient; Acupoint catgut embedding therapy can increase the ovariectomized rat uterus, the organ coefficient of the hypothalamus, to improve the organ's aging, the degradation of the situation. |
