| Objective The purpose of this study was to evaluate the effects of posphonized chitosan membrane in guiding tissue regeneration, and to provide the basis for posphonized chitosan membrane on the therapy of periodontal disease.Methods The second incisor teeth , the first and the second premolars of three dogs were used to create chronic periodontitis model by surgically placing silk ligaments and high sugar diet, then randomly assigned to one control group and two treatment groups .The one of the treatment groups was cured by guiding tissue regeneration with phosphonized chitosan membrane(A group),the other was cured by guiding tissue regeneration with drug-loaded phosphonized chitosan membrane(B group),and the control group was treated by flap operation only(C group) .The probing depth was evaluated through clinical examination before GTR and after the animals were killed. The regenerated tissue was observed through x-ray inspection .The animals were sacrificed at 12 weeks after surgery for histological analysis.The studied teeth with surrounding soft and hard tissues were harvested,repaired, fixed in 10% neutral formalin for 2 days,decalcified in mixed acid for 60 days,trimmed,dehydrated,and embedded in paraffin,stained and measured histological.Results 1. probing depth :The experimental groups resulted in statistically signi-ficant reduction in periodontal probing depth(P<0.05),when compared with the co-ntrol group.2. Radiography: The experimental group show that the formation of n-ew bone after 12 weeks .3. Histological measure :The amount of new alveolar bone formation, new cementum formation, new connective tissue adhesion in t-he A group was (1.22±0.15)mm,(1.00±0.15)mm,(1.72±0.17)mm, respectively. The B group:(1.44±0.14)mm,(1.20±0.19)mm,(2.27±0.18)mm; the C group:(0.56±0.16)mm,(0.51±0.14)mm,(1.27±0.16)mm. New periodontal tissue formation in the experi-mental groups were significantly higher than that in the control group .The diffe-rence was also significant between the two experimental groups(P <0.0 5).Conclusion Phosphonized chitosan membrane can be used as a biological material for GTR,and the effect of promoting periodontal tissue regeneration was more obvious after drug-loaded. |
