Background:Periodontal disease is one of the main causes of adult tooth loss.Oral diseases such as periodontal disease will cause bone tissue defects and eventually lead to the loss of oral functions.In recent years,tissue reconstruction has been achieved through surgery using barrier membrane.In the process of reconstruction,guided tissue regeneration(GTR)and guided bone regeneration(GBR)make use of the barrier function of membrane,so that fibroblasts in this area will not invade the bone defect,and then affect the growth of bone tissue.In this study,polycaprolactone(PCL),polyvinyl alcohol(PVA)and tea polyphenol(TP)were blended,and the nanoscale guided tissue regeneration/guided bone regeneration membrane with biocompatibility,barrier function,appropriate mechanical properties and biodegradability was prepared by electrospinning,and it was endowed with antibacterial and osteogenic characteristics.Objectives:1.PCL/PVA and PCL/PVA-TP fiber membranes were prepared by electrospinning,and their barrier function,mechanical properties and degradation behavior were investigated after characterizations.2.The experiments in vitro confirmed the antibacterial ability of PCL/PVA-TP.And explore its biocompatibility and osteogenic ability.3.The experiment in vivo confirmed that PCL/PVA-TP has the ability to promote bone regeneration under the condition of skull defect in rats.Methods:1.PCL,PVA,PCL/PVA,PCL/PVA-TP(PCL/PVA-TPL,PCL/PVA-TPM and PCL/PVA-TPH)were prepared and characterized by scanning electron microscope,Fourier transform infrared spectrometer and contact angle analyzer to confirm the synthesis of the composite.2.Through the barrier function and mechanical properties,the properties of PCL/PVA and PCL/PVA-TP were analyzed and compared,and the best ratio of PCL/PVA-TP was screened out.Select the best proportion to explore its degradation behavior.3.The antibacterial activity of PCL/PVA-TP was investigated by inhibition zone and absorption method,the cell compatibility of PCL/PVA and PCL/PVA-TP was evaluated by cytotoxicity test in vitro,and its osteogenic effect in vitro was also evaluated.4.Establish a rat skull critical size defects model,and evaluate its ability to promote bone regeneration by Micro-CT and histological staining after implanting materials.Results:1.The construction of PCL/PVA and PCL/PVA-TP electrospinning membranes was confirmed by characterizations.2.Through the barrier function test,it was confirmed that PCL/PVA and PCL/PVA-TP have macroscopic barrier properties that they hardly contract under the condition of 37 ℃ body fluids.However,PCL/PVA-TPL and PCL/PVA-TPH of PCL/PVA-TP were not closely attached to tissues.The evaluation of mechanical properties showed the excellent mechanical behavior of PCL/PVA and PCL/PVATPM.Degradation behavior is suitable for tissue growth process.4.Both inhibition zone and absorption method showed that PCL/PVA-TPM had a good inhibitory effect on the growth of Staphylococcus aureus and Escherichia coli.MC3T3-E1 was inoculated on the fibrous membrane,and its biocompatibility was performed well.Compared with PCL/PVA,PCL/PVA-TPM had higher alkaline phosphatase activity and the number of calcium nodules,and it had the ability to promote bone differentiation.5.PCL/PVA-TPM could obviously promote bone regeneration in the rat skull critical defect model.Conclusions:In this study,a bioactive nanofiber membrane PCL/PVA-TP was prepared,which has biocompatibility,barrier function,mechanical properties and biodegradability,and could promote osteogenic differentiation and antibacterial effect.It showed the application prospect in guided bone/tissue regeneration.
