The Effect Of Xuefuzhuyu Decoction On The Rats's Aortic Rings New Vessels

Xuefuzhuyu Decoction can improving blood high-coagulate state of tumor sufferers, preventing tumor emboli growing, killing tumor cells in blood circulation which break away from original tumors. However some researchers believe that medicines for blood-activating and stasis-dissolving are profitless for tumor because of the facilitating effect on the metastasis of tumors.In this paper, we studies the effect of representative traditional Chinese medicine Xuefuzhuyu Decoction on the neovascularization, finding out whether the Xuefuzhuyu Decoction facilitating or inhibiting angiogenesis.The study using aortic ring culture model .At first, we studied the culture methods. Then decided the way of culturing aortic rings is:1) The concentration of thrombin is 100NIH u/mL, the volume ratio of thrombin and fibrin is 6:1.2) Aortic rings were cultured in the 48 well plates. The volumes of tray gels and coverage gels is 200ul.3) The aortic rings were cleaned in PBS,and cut in 1mm. The total trimming time is no more than 2 minutes.4) Each well added 500ul medium. Xuefuzhuyu Decoction were added into the medium at the propotion of 5%,10%,15%,20%. Then observing its influnces on the endothelial cell, decided the Xuefuzhuyu Decoction whether promoting or inhibiting the angiopoiesis. The experiment set three concentration gradient, 0.5g crude drug/ml, 1g crude drug/ml, 2g crude drug/ml. To avid false positive phenomenon, the study using drug contained serum. Active positive control group added Endostatin.The immigration areas, IOD value, gray vales are selected as evaluation indexes. The results shows that: 1) The endothelial cell immigrating areas, IOD values, the grey values are all differently different between the sample groups and control groups. (P<0.05). 2) When the concentration and volume of Xuefuzhuyu Decoction increasing, the inhibiting efforts intensificated definitely. 3) The drug contained serum group shows that the Xuefuzhuyu Decoction really can inhibit the endothelial cells proliferation and immigration, and there is no false positive phenomenons exited in this experiment.Conclusion:1) To sum up the classic traditional Chinese medicines Xuefuzhuyu Decoction which effects on blood-activating and stasis-dissolving have an inhibitory effect on endothelial cells' proliferation and immigration.2) The inhibitory effects amplified gradually when the concentration and volume of Xuefuzhuyu Decoction increasing.
