Study On The Anti-Tumor Activity Of Typhonium Lotus Tuber Extraction

Typhonium lotus is a kind of Chinese traditional medicine. It has high medical value and function of prevents disease. The extraction of typhonium lotus tuber was used to study its antitumor activity and discussed its related mechanism.Use the tuber of Northeast typhonium lotus as material, with 10 times of distilled water as extraction solvent, extraction methods is using ultrasonic, the time is 20min, one repeat after lmin interval, the extraction of typhonium lotus tuber is obtained after vacuum drying. It's the best relative inhibition rate on tyrosinase is 51.21%. The results of MTT show that the extraction of typhonium lotus tuber has some certain inhibition on the proliferation of Hela, and inhibition rate is 65.08%. Apoptosis and flow cytometry analysis show that the extraction of typhonium lotus tuber can cause the increase of apoptotic cells, and this phenomenon is dose-dependent. The apoptosis rate is the highest after treated with 72h by the extraction of typhonium lotus tuber, reach to 24.4 %.Western blot detect Hela cells was treated 48h by a series of concentration of extraction, the expression of caspase 3 was increased gradually. It appeared good dose-effect relationship about the extraction of typhonium lotus tuber on cancer cell inhibition. The extraction of typhonium lotus tuber on the mice with solid tumor by inocula S180 cell line show that anti-tumor activity of typhonium lotus is significant, the inhibition rate reach to 31.77%. Experiment of extraction of typhonium lotus tuber that gavaged to Kunming mice with ascites carcinoma by inocula EAC cell lines shows that: the extraction can extend survival time, the extended rate reach to 43.26%, and can improve the survival state of mouse with ascites carcinoma. Results reveal the extraction of typhonium lotus tuber has distinct anti-tumor activity.
