Ferula sinkiangensis K.M.Shen, a useful Chinese special medicinal plant which at the edge of extinction, was selected as the research material. Present study based on the seed embryo germination and culture,and provided a feasible approach for protecting and developing the resources of Ferula sinkiangensis K.M.Shen. The reason of seed dormancy , autotoxicity effects, suspension culture and hairy root induction were studied in this Paper . The main results were summarized as following :1 . W e can conjecture that the effector site of seed dormancy may exist in seed coat .the seed embryo germination and culture :results showed that the optimum medium for embryo culture was MS( pH 5.5~6.0)+6-BA 0.1 mg/L + GA31 mg/L +3% sucrose; different germination temperature was prominent for germination, the optimum was 20,25,30℃.2 . Germination inhibitors and seed dormancy : germination inhibitors in whole seed highest amount is in seed coat, which main restrain seed germination and induce seed dormancy of Ferula sinkiangensis . The site of cold stratification breaking dormancy may exist in whole seed except for seed embryo. Cold stratification do harm to embryo , but make germination inhibitors degrade or converse. The effector andinhibitor co-exist in seed ,however ,the interaction mechanisms of both remain to be study.3 . Shoot could be induced in MS medium , contained with IAA0.1 mg/L,NAA 1.0mg/L,6-BA0.5 mg/L ; suspension culture and hairy root : The medium for calus inducing was MS +0.4mg/L2,4-D+1.0mg/L 6-BA+1.0KT mg/L. Agrobacterium rhizogenes-mediated root transformation was established.4 . Autotoxicity effects : in present study the potential autotoxic effect of aqueous extracts from Ferula sinkiangensis seed was investigated. At high concentration, the seed embryo germination and seedling growth was significantly inhibited ,while with concentration decreasing ,the inhibitory effect got weaker . Higher concentration of extracts stimulated creation the MDA and peroxidation of membrane lipids. The results showed that autotoxicity was partially responsible for natural regeneration of Ferula sinkiangensis.
