| Cervical cancer is the second most.frequently occurred tumor in female reproductive system worldwide and its incidence rate is rapidly increasing among young women every year.So its threat to health can not be ignored.A lot of evidences have displayed that the cervical cancer is the only tumor whose rates of incidence and mortality can be decreased by means of medical interference.So it's essential to find a new effective drug target for clinical treatment as soon as possible.In 1995,through epidemiology examination in 22 countries' patients,establish the infection of human papillomavirus(HPV) plays an important role in the carcinogenesis and development of cervical cancer.However,the infection of HPV is generally among people,only a few females with HPV infection have developed cervical cancer,which suggests that if there are other factors involved besides HPV infection or not need us advanced research.The Notch-Hes signaling pathway plays an important role in the process of cell proliferation,differentiation and apoptosis.Notch-1 and Notch-2 belongs to important signaling receptor protein family in development of vertebrate and invertebrate.They affect tumor cell growing,apoptosis and blood vessel formation by cooperation, complemention or antagonism way.Until now,there have been few researches about Notch-1 and Notch-2 in cervical cancer.As an important target gene,whether Hes gene expression or not is the marker of Notch signaling pathway activation.The research about the relation between Hes and human cervical cancer is few,and in different tissues Hes plays different roles.Generally acknowledged that P16 is an anti-oncogene, its expression is correlated HPV quantity,we can elevate the sensitivity and specificity of diagnosis and the accurate of judgement prognosis in cervical cancer.Here,we have examined the expression of Notch-1,Notch-2,Hes-1 and p16 in both human cervical cancer tissue and normal cervical tissue,to approach their role in cancer development, and provide us a new strategy for tumor therapy.MethodWe make tissue array to examine the expression of Notch-1,Notch-2,Hes-1 and p16 in both human cervical cancer tissue and normal cervical tissue by immunohistochemistry,and their mechanism of action in cervical cancer development.ResultThe expression of Notch-1 in normal cervical tissue is only 5.88%,it is higher in squamous cell carcinoma of cervix and cervical adenocarcinoma tissue(46.43%, 59.38%).There are significant differences of expression of Notch-1 among normal and cervical cancer tissue(P<0.005).The expression of Notch-2 in normal and cervical cancer tissue show no difference,such as 76.47%,78.57%and 81.25%(P>0.05).The expression of Hes-1 in these tissues is 64.71%,96.43%and 93.75%,there are also significant differences of expression among normal and cervical cancer tissue(P<0.05). The expression of p16 is average among normal,squamous cell carcinoma of cervix and cervical adenocarcinoma tissue,they are 76.47%,82.14%and 93.75%,and no significant differences(P>0.05).The expression of Notch-1 and Hes-1 in cervical cancer tissue is higher in normal tissue(P<0.05);The expression of Notch-2 and p16 have no significant differences(P>0.05).The expression of four proteins in squamous cell carcinoma of cervix and cervical adenocarcinoma tissue have no significant differences(P>0.05),but all of them are high.Conclusion1.The expression of Notch-1 and Hes-1 may be associated with the carcinogenesis and development of cervical cancer,the key point may be the Notch-Hes signaling pathway,it may be a new direction in cervical cancer therapy pointing on the Notch-Hes signaling pathway;Notch-2 is only participate cell hyperplasy;2.The expression of p16 in cervical cancer and normal cervical tissue is high,witch proved that maybe there have HPV infection and the Notch-Hes signaling pathway may play a synergistic effecting with HPV in the carcinogenesis and development of cervical cance. |
