| Object: To study the expression of Notch-1 in human gliomas and the correlation between the expression and the pathological grades of gliomas, to explore the role of Notch-1 in the tumorigenesis and progression of human gliomas, accordingly to provid some proof of theory for treatment of human gliomas.Method: The expression of Notch-1 mRNA and Notch-1 protein was detected in 70 primary human gliomas and 12 normal cerebral tissuses by using RT-PCR semi-quantity and Western Blot methods; immunohistochemistry was used to examine the expression of Notch-1 protein in 83 primary human gliomas and 12 normal cerebral tissuses.Results: The expression of Notch-1 mRNA and Notch-1 protein have been detected in human gliomas and normal cerebral tissuses and the expression level of Notch-1 mRNA and Notch-1 protein in human gliomas was considerably higher than those in normal cerebral tissuses(P<0.01); the expression level of Notch-1 mRNA and Notch-1 protein in gliomas of grade I~II and III~IV was considerably higher than those in normal cerebral tissuses(P<0.01),and the expression level of Notch-1 mRNA and Notch-1 protein in gliomas of grade III~IV was considerably higher than those in gliomas of grade I~II.Conclusion:1.The expression of Notch-1 have been detected in human gliomas and normal cerebral tissuses, and the expression of Notch-1 in human gliomas was... |
