The Experimental Study On Effect Of Nuanxin Capsule On Preventing And Treating The Congest And Its Mechanism

ObjectiveAccording to the theory of traditional Chinese medicine the Professor Deng Tietao treats Congest Heart Failure,the deficiency of heart and kidney.Based on this pattern of heart and kidney deficiency,this study explores pharmacological mechanisms of Nuanxin capsule-the empirical prescription of clinical treatment of Congest Heart Failure of Professor Deng Tietao,In this study,according to Professor Deng Tietao"heart failure based on the Yang Qi-deficiency"theory,to explore the pharmacological mechanism of Nuanxin capsule.By observing the effects of Nuanxin capsule on hemodynamics index,serum superoxide dismutase,myocardial cell apoptosis and the weight of heart and pathological changes,to explain the Nuanxin capsule's target for the treatment of heart failure.To promote the clinical use of Nuanxin capsule to treat the Heart Failure.1 Theoretical StudyAbout the research on the heart failure,in recent years,most of medical experts think that as a result of energy shortage or the imbalance of energy metabolism,myocardial cells'struction and function damages.The overloading and metabolic disease happens on heart.AngiotensinⅡand free radicals plays the very important role in the reconstruction of heart.When the heart failure happened,LPO and MDA increase obviously,the synthesis of SOD blocked.As a result,the scavenging ability of OFR decreased obviously.2 Experimental Study Experiment 1:experimental study of effect of Nuanxin capsule on rats Heart Failure induced by abdominal aorta constriction.Rat models of heart failure(HF) were induced by the method of abdominal aorta constriction.Then the HF rats were randomized into the model group.Nuanxin capsule(NC) groups(treated with NC at 0.825 and 1.650 g·kg^-1d^-1 respectively),and digoxine group(digoxine at the dose of 0.091g? kg^-1d^-1).Meanwhile,the pseudo-operation group was established.After treatment for 30 days,the heart rate,systolic blood pressure(SBP),diastolic blood pressure(DBP),left ventricular systolic pressure(LVSP),left ventricular end-diastolic pressure(LVEDP),maximal rate of increase and decrease in left ventricular pressure(±dp/dt) as well as levels of serum superoxide dismustase(SOD) and malondialdehyde(MDA) were measured.Experimental 2:experimental study of effect of Nuanxin capsule on Heart Failure rat model induced by adriamycin.Forty SD rats were used for the study and heart failure was induced by intraperitoneal injection of adriamycin.The survival rats were randomly divided into 4 groups:model control group,middle-dose and high-dose NC groups(0.825,1.65g/kg) and digoxin group(0.091g/kg).Each group has 10 rats.Another 10 healthy rats were in the normal control group.Except the normal control group and model group received gastric infusion of equivalent distilled water,the other groups were given the corresponding drugs everyday for 30 days.After the administration for 30 days,the serum levels of angiotensinⅡ(AngⅡ) and superoxide dismutase(SOD),and the rat heart rate were examined,the heart coefficient was calculated,and the heart size was measured.Experiment 3:experimental study of effect of Nuanxin capsule on the hypoxia ability of rats Rats are randomly divided into four groups,namely the control group;Nuanxin capsule of the low-dose group,;Nuanxin capsule of the high-group;positive group.Gives drugs 7 days.After 7 days,conducts esperiment.To observe the effect of the impact of hypoxia on rats.Results:Experiments 1:High dose NC and low dose NC increased SBP;LVSP and±dp/dt,decreased LVEDP,increased SOD activity,and reduced MDA content.The difference being significant as compared with the model group(P＜0.05).NC also slowed heart rate,but the difference was insignificant(P＞0.05 compared with that in the model group).Experiment 2:NC could reduce the heart rate,and decrease the heart size and heart coefficient of heart failure rats,and its effect was similar to the positive control drug.Middle-dose and high-dose NC significantly reduced serum AngⅡlevel of heart failure model,and high-dose NC increased the serum SOD activity obviously.Experiment 3:In some degrees,Nuanxin capsule can extend the atmospheric hypoxia survival time of rats.Nuanxin capsule of the high-dose have better effect than the low-dose.Conclusion:The therapeutic mechanism of NC for heart failure is probably related with the improvement of cardiac systolic and diastolic function,and with the regulation of free radical metabolism and the inhibition of myocardial lipid peroxidation.Nuanxin Capsule has a good therapeutic effects on the rats models of adriamycin-induced heart failure.
