Objective:This study was to investigate the distribution of the gene polymorphism in FOXC2 C-512T and FOXC2 G-350T,to investigate the level of FOXC2 mRNA in ventral adipose tissue,and to assess the association of the FOXC2 gene with Type 2 Diabetes and Metoblic Syndrome(MS).Methods:The subjects were involved into DM+ and DM- groups,MS+ and MS- groups, investigated the polymorphism in FOXC2 by PCR-RFLP,investigate the level of FOXC2 mRNA in ventral adipose tissue by RT-PCR.Results: 1 The allele C of FOXC2-512 correlated with MS and obese,but the polymorphism didn't significantly correlated with T2DM.The T allele of FOXC2-512 correlated with the lower BMI,waistline,hipline,WHR and fat%. 2 The polymorphism of FOXC2 G-350T didn't correlated significantly with T2DM and MS. 3 The groups with DM+ or MS+ had lower level of FOXC2 mRNA in visceral adipose tissue. The level of FOXC2 mRNA correlated negatively with BMI,HOMA-IR and blood pressure. 4 The level of FOXC2 mRNA in visceral adipose tissue didn't correlated significantly with the gene polymorphism in FOXC2 C-512T and FOXC2 G-350T.Conclusion:The allele C of FOXC2-512 correlated with MS and obese,but the polymorphism didn't significantly correlated with T2DM.The groups with DM+ or MS+ had lower level of FOXC2 mRNA in visceral adipose tissue.
