| Objective:To observe the JianPi ShuGan JieDu(JPSGJD)recipe to the evoked model of liver cancer cells in rat,explore its preventive and therapeutic effect and the mechanism,with a view to effective clinical use of the JPSGJD recipe detoxification of liver cancer,especially provide an objective basis to the earlier period of the hepatoma.Methods:SPF-SD male rats,six weeks old,were divided into 6 groups:To establishe the DEN control group,Low-dose of group,High-dose of group,Model group,High-does of Model group,Low-dose of Model group.And then,the control group was interruptable 95μg/l DEN,model group was produced the splenic asthania,stagnation of liver-QI and cancer by interruptable 95μg/l DEN lucuriate ange and rhubarb,High-dose of group and High-dose of Model group are given medicine according to 30mg/kg,Low-dose of group and Low-dose of Model group are given medicine according to 10mg/kg,Control group and Model group are given physiological saline,and all of them are medicated administrtion team for 150d gavage;and we check the weight and action in this period.The day after the end of the experiment,the rats are killed and weighed the liver,the spleen and the weight.Calculated the rats of inhibirory effect of liver carcinoma and the index of the spleen.HE stain,light microscopy level of liver cancer,We measuered the expression of c-myc mRNA and p16 mRNA through the real-time fluorescence quantitative Riverse Transcription-Polymerase Chain Realtion(RT-PCR).To explore the mechenism of the JPSGJD recipe on the liver cancer of the causality.Results:At the end of all of the experiments,the rats in the Control group move slowly,decrease the drink and food,and their sedes are thin,their pelage are neat;the rats which the pelage,feeling,eating and drinking in the High-dose of group are normal;the rats in Low dose of group are slightly normal,but their eating are reduced,thier sedes are slightly thin,and the pleges are unnormal;Model-group are less excitement,emotional lability,their body weight are decreased,and anus are dirty;the High-dose of Model group is common,their sedes are formed,and pelage are inmarsh;the Low-dose of Model group less move,emot unsteadly,less eat,their belages are more better;The Control group's orgnization of lobuli hepatis are distroyed and displaced by nothogolial,hyperplastic fibrous tissue divided virgin lobuli hepatis,and circumvolutio inequality of size hepatic cell,the central veins absence can be seen,a little chromatospherite are incement,and the quantities ate increased obviously,inflammatory cell infiltrations in portal area are increased slightly,fibrosis which are penim the lobuli hepatis are more obviously,the High-dose of group are decreased obviously than Control group,the fibrosis near the lobuli hepatis;the Low-dose of group are slightly relieve than Control group;The hepatic cells in Model group are disorder and some of them are vaculization,necrosis,the size are different,the shape is unnormal,the cellular nucleus are big,some hepatic cell is big and a heteromorphism,the blood vessel hyperplasia is aggravate;The hepatic cells of High-dose of Model group are disorder,the cells are necrosis,hyperplasy and phlegmasia in different degree;The hepatic cells of Low-dose of Model group are more serious than the High-dose of Model group in the aspects of necrosis and hyperplasy;It has showed out that the JPSGJD recipe has conspicuous prenentive and theapeutic effect.The spleen index of the High-dose of group is increased clearly compared with the Control group(P＜0.01);There is statistical significance between the spleen index of the Low-dose of group and the Control group(P＜0.05);The liver index of the Control group is increased clearly compared with the High-dose of group and the Low-dose of group(P＜0.01);The expression of c-myc mRNA and p16 mRNA is different between Control group,High-dose of group and Low-dose of group;There is statistical significance in each other(P＜0.01);The spleen index and the liver index of the High-does of Model group is increased clearly compared with the Model group(P＜0.01);The expression of c-myc mRNA in Model group is different from High-does of Model group(P＜0.01),and have statistical significance compared with Low-dose of Model group(P＜0.05);The expression of p16 mRNA is different between the Model group and High-does of Model group(P＜0.05)。Conclusion:The JPSGJD recipe has conspicuous prenentive and theapeutic effect.Its mechanism of action may be related to reduce the expression of c-myc nRNA and inhibit the devitalization of p16 mRNA.It is easy to induce cancer under the tagnation of liver-QI with deficiency of the spleen stimulating factor.It also can remarkably inhance the rat's immune function. |
