Objective:To investigate the concentration and distribution of alkaline phosphatase(ALP) in gingival tissues of experimental periodontitis in rats and its correlation with periodontitis.Methods:Experimental periodontitis in rats was produced, then the rats sacrificed respectively after 4 weeks and 8 weeks. The ALP concentration was assayed by microplate reader;the Gomori's method was used to make histological section, and the distribution of the ALP was observed by optical microscope.Results:1. The variance of the ALP concentration in gingival tissues.ALP concentration in gingival tissues was significantly higher in periodontitis group at four and eight weeks after operation than that in the control group(P<0.01);in periodontitis group, the ALP concentration in gingival tissues was significantly higher at eight weeks than that at four weeks after operation(P<0.01). 2. The distribution of the ALP in gingival tissues.The dense of ALP in gingival tissues was significantly higher in periodontitis group at four and eight weeks after operation than that in the control group;in periodontitis group, the dense of ALP in gingival tissues was significantly higher at eight weeks than that at four weeks after operation. Conclusion:In periodontitis group periodontal lesion was appeared at four weeks after operation, Alveolar bone loss was found in mesio-distal direction and furcation from ligated teeth. And ALP concentration in gingival tissues was significantly higher in periodontitis group than that in the control group. The degree of periodontal lesion was more obviously at eight weeks after ligation, Alveolar bone loss was more significant from ligated teeth and adjacent teeth. And ALP concentration in gingival tissues was obviously higher than that in the four weeks group. Therefore, in this experiment ALP concentration in gingival tissues was significantly higher in periodontitis group than that in the control group. And ALP concentration was obviously higher with the progress of periodontitis. So, the concentration of ALP in the gingival tissues are related to the degree of periodontitis.
