ObjectiveAs an important primary chemical material, vinyl chloride is mostly absorbed through respiratory system and generates carcinogenesis, mutagenesis and teratogenesis. It was reported that vinyl chloride could impair the genital system in 1970s.But it has not arrived at a consistent conclusion. In the present study, the occupational vinyl chloride-exposed workers were selected.Using the laboratory detecting and epidemiological investigation to find whether it has potential effects on the reproductive toxicity to them which have occupational vinyl chloride exposure.MethodsSubjectExposed groups:The occupational vinyl chloride-exposed workers were chosen as exposed group and divided into low-level exposure group and high-level exposure group according to the levels of vinyl chloride in workplaces and in individuals.Controls:In the same city, the non-exposed workers in machining and carborundum workshop were chosen as control group.Methods1.Epidemiological investigationAll of the item about the questionnaire will be asked and filled in by the uniform trained investigators.Collect the exposure-gas both of the workplaces and individual contacted.2.Laboratory determinationThe empty stomach venous blood of all of subjects were collected and centrifuged. The levels of FSH,LH,InhB,T,E2 in serum were then evaluated by the enzyme-linked immunosorbent assay kit(Adlitteram Diagnostic Laboratories). The OD value was determined by Microplate reader at theλ450nm.The collected gas was determined by vapor-phase chromatography.3.Statistics MethodsAfter checking, the data were entered with Excel. All the analyses were carried out using the SPSS 13.0 software. Results were expressed as means±SE and rate. Compare with the control:The distribution of measurement data was examined first, and then the data was analyzed by using t-test or t'-test according to the distribution. The difference among multiple groups was done by one-way analysis of variance (ANOVA),the difference between two of them was done by SNK-q test when F was significant. The relevance of data was analyzed by using trend x2 and linear regression. P-values of less than 0.05 were considered statistically significant.Results1.In the male exposed group, the concentration of InhB is 342.82±6.32pg/ml, which is significant higher than the control(p<0.05).The concentration of T and E2is 7.33±0.51ng/ml and 222.80±23.41pg/ml,respectively,which is significant lower than the control(p<0.05).2.There is a negative correlation of the concentration of T in male and the levels of vinyl chloride exposure dose, and the correlation coefficient is -0.264(p=0.004).The regression equation is T=9.731-0.43ρ(T:testosterone,ρ:the vinyl chloride exposure dose,(P<0.05).3.The spontaneous abortion rate of the vinyl chloride high-level exposure male workers' wives is 10.7%,which is significant higher than that of low-level exposure and control groups(0.0%) and that of Liaoning Province(3%-4%).The rate of the low-level exposure female workers is 12.5%,but the rate of other groups is 0.0%. There is no significant difference in the female groups. The spontaneous abortion rate of the couple that both exposed to the VC(12.5%) is much higher than that of only the husband exposed to the VC(3.3%,P<0.05).4.There is a statistical significant among the spontaneous abortion rate of the vinyl chloride exposure male workers' wives, the vinyl chloride exposure dose and time through trend-x2 analysis. There is a significant positive among the spontaneous abortion rate, the exposure dose and time, and the correlation coefficient is 0.213,0.210,respectively(p<0.05).ConclusionVinyl chloride exposure can affect the reproductive hormone levels in workers, and increase the spontaneous abortion in workers' wives. So,vinyl chloride can affect the reproduction system of VC-exposed workers. There is the trend increasing with occupational exposure time.It is more prone to spontaneous abortion to the woman exposed to vinyl chloride with her husband exposed, either.
