To provide experimental basis for clinical application.2.Methods60 male Wister rats were randomly divided into six groups, followed by a blank group, high dosage group, middle dose group, low dosage group,Dingchuan Decoction group, model group,every group has 10 rats. Asthma model with reference to the introduction of equality Lu Ways, Divided into two steps sensitized and challenged. Sensitized: blank group with saline, the remaining groups with 1ml saline liquid preparation of OVA antigen (100mg egg albumin and aluminum hydroxide powder 100mg). There was difference of weight between the blank group and other groups before and after provocation, the weight of normal rats was higher than that of rats in other groups (p<0.01). There was no statistical significance among the groups (high dosage, middle dosage and low dosage of Sang Su Er Chen Tang Modified, Ding Chuan Decoction group and model group)(p>0.05).5.1.2 symptoms of rats in all groups at the time of serum provocationNormal rats were full of vigor and had prompt response, steady breath and smooth and clean fur. Shortly after inhalation of OVA, model rats came forth symptom of hypersensitiveness such as restless, scratching, licking limbs, sneezing and other allergic symptoms; then prostrated them without movement and were in tachypnea or dyspnea (visible abdominal breathing) with slight cyanosis. Some of model rats sounded wheeze during expiratory phase, mucosal fluid flowed from their mouths and noses and breathed irregularly. Repeatedly stimulated rat dull coat, lying tropic response lazy and slow responding.Compared to model group, symptom of Sang Su Er Chen Tang Modified high dosage group,middle group decreased, but remained tachypnea or dyspnea accompany with slight cyanosis, some of the rats sounded sonorous wheeze during expiratory phase and breathed irregularly. Sang Su Er Chen Tang Modified low dosage group and Ding chuan Decoction slightly shortness of breath, no cyanosis, wheezing and other symptoms soundCompared among high dosage group, middle group and low dosage group, symptoms of low dosage group improved dosage, with slighter tachynea or dyspnea. The fur of the rats was more smooth and clean with more active spirit.5.2 changes of bronchial and pulmonary histopathologyCompared with the blank group:bronchial epithelium and tracheal wall in model group thickened,Epithelium was damaged and shed significantly, narrow lumens could be observed; Beaker cell proliferated, mucus plug blocked the lumens; infiltration of considerable acidophil leukocytes and lymphocytes occurred around the trachea in large or middle size and blood vessels, as well as slight hyperplasia in bronchial and smooth muscle, increase of phagocyte and widening of alveolar septum.Compared with the model group: bronchial lumens in Sang Su Er Chen Tang Modified low dosage group and Ding chuan Decoction were almost normal with slight hyperplasia of epithelium; no obvious narrow lumens and inflammation in tracheal wall and a few beaker cells could be observed; Bronchial around a small amount of lymphocytic infiltration, as well as slight hyperplasia in bronchial and vascular smooth muscle; Compared to model group, the decrease of phagocyte and slightly widening of alveolar septum were significant.Phase comparison, pathological inflammation of Sang Su Er Chen Tang Modified high dosage, middle dosage group were slighter than that in the model group, but more severe than that in Sang Su Er Chen Tang Modified low dosage and Ding chuan Decoction group, obvious tracheospasm, inflammatory excretion in secretions lumens and obvious narrow lumens could be observed.Among the three dosage group of Sang Su Er Chen Tang Modified, the effect of low dosage was better than that of high dosage and middle dosage, the effect of low dosage is best which showed slightest inflammation in air passage. A few infiltrated inflammatory cells could be observed in lumens of middle dosage group.The thickening of bronchial wall in high dosage group, hyperplasy and edema in epithelium, considerable acidophil leukocytes and lymphocytes occurred around the trachea in large or middle size and blood vessels.5.3changes of contents of cytokinesCompared with the blank group, the content of asthma model group rats ET-1, IL-1,IL-6,TNF-α,No,were significantly increased, lung tissue`s epithelial cell ICAM-1 expression markedly increased, suggesting the existence of asthma inflammation. With the asthma model group, both treatment groups to alleviate inflammation, ET-1, IL-1, IL-6, TNF-α, No, content decreased, lung tissue`s epithelial cell ICAM-1 expression reduced. With Ding chuan Decoction group, Sang Su Er Chen Tang Modified high, meddle dose group ET-1, IL-1,IL-6,TNF-,No content increased (p<0.01), lung tissue`s epithelial cell ICAM-1 strong expression (p<0.01);and Sang Su Er Chen Tang Modified low dosage group without statistical significance. Sang Su Er Chen Tang Modified Senior low dosage group, low dosage high, intermediate amount of ET-1,IL-1,IL-6,TNF-α,and No decreased (p<0.05,p<0.01),lung tissue`s epithelial cell ICAM-1 reduced expression (p <0.01).Conclusions:1.In the lung tissue morphology, Sang Su Er Chen Tang Modified low dosage group can accelerate to alleviate bronchospasm and reduce eosin Phil and lymphocyte infiltration, reducing airway inflammation, which diffusing the lung to resolve phlegm and clearing away heat to relieve asthma drug (governance such as feathers on coke, non-light does not cite) has provided an effective basis for the treatment of asthma.2.Diffusing the lung to calm panting and clearing away heat to resolve phlegm can significantly improve the serum TNF-α, ET-1, IL-1, IL-6, NO content and lung tissue`s epithelial cell ICAM-1 positive expression area of effective control over gas airway inflammation, thereby improving airway remodeling.3.Experimental acute attack prompted the use of diffusing the lung to resolve phlegm and clearing away heat to relieve asthma drug effect, we must pay attention to light dosage, Do not have to use cold. |
