Experimental Study Of San’aoxiongting Mixture On Neurogenic And Immune Airway Inflammation In Asthmatic Rats | Posted on:2011-09-04 | Degree:Doctor | Type:Dissertation | Country:China | Candidate:Y Jing | Full Text:PDF | GTID:1484303323954079 | Subject:Chinese medical science | Abstract/Summary: | PDF Full Text Request | Objective:by observing the influence of san’aoxiongting mixture on the expression of NGF、CGRP、SP、IFN-γ、IL-4 and IFN-y/IL-4 of serum in asthmatic rats, this paper aims to explore preliminarily the function mechanism of san’aoxiongting mixture on asthmatic airway inflammatory.Methods:The 100 health SD rats were randomly divided into six groups—normal group, asthma model group, high dosage group, middle dosage group, low dosage group and dexamethasone group. Except for the normal group was made as model with physiological saline, the other five groups were made models with OVA and aluminum hydroxide. Since the beginning of the third week, each group excitated with physiological saline (normal group) or 1%OVA (the other five groups), and separately fed with physiological saline (normal group and model group), san’aoxiongting mixture (high, middle and low dosage), or abdominal injected with dexamethasone (dexamethasone group) one hour before every excitation. All rats were killed at the end of the third week. The airway inflammation and Structural Change were examined by microexamination; expressions of CGRP、SP、IFN-γ、IL-4 in lung were detected by immunohistochemical method respectively; in the mean time, expressions of NGF and NGFmRNA in lung were measured by immunohistochemical method and as well as the in-situ hybridization method; the levels of IFN-y/IL-4 of serum were examined by the enzyme linkedimmunosorbent assay.Results:compared with the normal group, the numbers of eosinophils and lymphoeytes、the thicknesses of WA/Pi and ASM/Pi and also the expressions of CGRP、NGF、NGFmRNA IL-4 in lung and the levels of IL-4 of serum in model group were all higher; the expressions of IFN-γ、the levels of IFN-y of serum and the ratio of IFN-y to IL-4 were all lower; The numbers of eosinophils and lymphoeytes in san’aoxiongting combined group and dexamethasone group were much more lower than model group; In addition, The thicknesses of WA/Pi and ASM/Pi in san’aoxiongting high dosage group, middle dosage group and dexamethasone group all were lower than those of the model group and the expressions of CGRP、NGF、NGFmRNA in lung in san’aoxiongting combined group and dexamethasone group were much more lower than the model group; Moreover, in comparison with the model group, san’aoxiongting combined group and dexamethasone group were much more lower in expressions of IL-4 and the levels of IL-4 of serum; and the expressions of IFN-y together with the ratio of IFN-y to IL-4 were both significantly higher in san’aoxiongting combined group and dexamethasone group than in the model group.Conclusions:san’aoxiongting mixture can reduce the airway neuroimmune inflammation by down-regulate the expressions of NGFmRNA, hence decrease the expression of neuropeptide substance in lung tissue, probably further correct the imbalance of Th1/Th2;Besides it also can alleviate the asthmatic airway remodeling and partly restore of appropriate structure of airway wall.
