| In this study, we reported five hES cell lines (J6,J13,J14,J30 and J32 ) being established from the Chinese population and characterized in a comparative way. The blastocysts used for embryonic stem cell derivation are clinically IVF surplus blastocysts. All the cell lines had morphological characteristics typical of hES cells. We had characterized two cell lines-J6 and J13. They expressed alkaline phosphatase activity and molecules typical of primate embryonic stem cells, including Oct-4, SSEA-3, SSEA-4, TRA-1-60 and TRA-1-81. They formed embryoid bodies that expressed markers of a variety of cell types. They formed teratomas with tissue types representative of all three embryonic germ layers. These human embryonic stem cells are capable of producing clones of undifferentiated morphology, and J6 and J13 cell lines were propagated to become sublines (J6-1 and J13-1) which still retained undifferentiated morphology in vitro. The PD time of the two newly established cell lines were 31.43h(J6) and 21.38h(J13). At present, J6, J13, J14, J30, J32 have reached approximately 485, 478, 259, 120, 121 days respectively. |
