Objective:Exploding the effect of the expression on TGF-β1 after injecting asiaticoside on Oryctolagus cuniculus model,in order to find mechanism of hypertrophic scar and provide a treating method.Methods:20 New Zealand whites rabbits were randomly divided into 4 teams:negative group,diprospan group,asiaticoside(25mg/ml) group and asiaticoside (50mg/ml) group.,and 6~8 scars(1cm in diameter on ventral side of rabbits ears)on rabbits' ears of 20 rabbits(four died ) were created in one rabbit,deeply to perichondrium.Corresponding drug was used 3 days later and once every other day,till to scars' preparation and specimen was made of 2 weeks later.Extractated the total TNA and measured TGF-β1 mRNA in the 4 groups by the meathod SYBR PrimeScript RT-PCR in order to find the difference.Results:Asiaticoside group has a lower expression of TGF-β1 mRNA compared with negative group,(P<0.05),while there are no respective diversity(P>0.05 ) between asiaticoside group and masculine group.As to the two asiaticoside groups,group of higher concentration has a lower expression of TGF-β1 mRNA and the difference is significant.Conclusions:Asiaticoside injected locally could inhibit the expression of TGF-β1 mRNA obviously,and the therapeutic effect has a direct proportion with drug concentration.Asiaticoside injected locally has positive effect in the treatment of hyperplastic scars. |
