Objective:To investigate the relationship between the expression of Survivin,Bcl-2 and Caspase-3 gene and the relationship among the expression of Survivin,Bcl-2and Caspase-3 gene with clinicopathologic factors in breast carcinoma.Meanwhile,their possible roles in the development and progression of breast carcinoma have also been studied.Methods:Streptavidin-biotin peroxidase(S-p) method of immunohistochemical staining was used to detect the expression of Survivin,Bcl-2,Caspase-3 in 10 benign breast tissues,10 atypical hyperplasia breast tissues and 56 primary breast carcinoma tissuesResults The positive rate of Survivin,Bcl-2,Caspase-3 in the primary breast carcinoma were 67.9%(38/56),62.5%(35/56),51.8%(29/56) respectively.In contrast,Survivin was negative in 10 normal breast tissues and 9 atypical hyperplasia breast tissues;there was a trend that from normal breast tissues to atypical hyperplasia breast tissues to primary breast carcinoma tissues,the expression of Bcl-2 became more higher.This trend was Statistieally significantly(x~2=10.118,P<0.01).And Caspase-3 was positively expressed in a significantly greater proportion of the tissues(100%) than breast carcinoma 51.8%(x~2=12.042,P<0.01).Survivin expression was relateted to lymph node metastasis (x~2=7.09,P<0.01),but not to age,tumor size,pathological grade and clinical stage(P>0.05).Bcl-2 expression was relateted to lymph node metastasis and pathological grade(x~2=8.44,P<0.01;x~2=8.26,P<0.05),but not to age,tumor size and clinical stage(P>0.05).No significance was seen in the Caspase-3 expression with age,tumor size,pathological grade and clinical(P>0.05).The expression of Survivin was positivly correlated with Bcl-2 in breast carcinoma tissues(r=0.317,p=0.017);The expression of Survivin was negatively correlated with Caspase-3(r=-0.405,p=0.02);The expression of Bcl-2 was negatively correlated with Caspase-3(r=-0.317,p=0.017).Conclusion:Survivin and Bcl-2 are over-expression while Caspase-3 is low-expression in breast carcinoma tissues compared with noncarcinoma tissues,the difference was significant,so there is apoptosis inhibition.Abnormal expression of Survivin,Bcl-2 and Caspase-3 is closely correlated with the carcinogensis and the development of breast carcinoma.Positive expressions of Survivin and Bcl-2 in breast carcinoma is positivly correlated.Survivin,Bcl-2 may play an synergetic role in the process of cacinogenesis of breast carcinoma.Both positive expressions of Survivin and Bcl-2 in breast carcinoma are negatively correlated with positive expressions of Caspase-3,This suggests that,In breast carcinoma,the inhibition of cell apoptosis is a result of their mutual action. |
