| Objective:To study the expressions and significance of B-FABP mRNA in tumor tissues and blood of patients with renal clear cell carcinoma(RCCC).Methods:B-FABP mRNA in tumor tissues of 55 patients with RCCC and 28 normal kidney tissues were detected by RT-PCR.RT-PCR-ELISA was used for testing the B-FABP mRNA expressions in blood of 55 patients with RCCC,and 28 healthy volunteers.Results:1.The positive expression rate of B-FABP mRNA in rumor tissues was 76.4% (42/55),which was notably higher than the 10.7%(3/28) expression rate in normal kidney tissues.The positive expression rates in clinical stages ofâ… ,â…¡,â…¢andâ…£were 57.1%,68.4%,93.6%and 100%respectively,and in pathological grades of G1,G2 and G3 were46.7%,80%and 100%respectively.The expression level of B-FABP mRNA was markly elevated as the clinical stage and pathological grade increased in RCCC. (p<0.05).The positive expression rate of B-FABP mRNA in patients with lymphatic metastasis was 100%,while 41.7%in patients without lymphatic metastasis,which showed significant difference with relationship to lymphatic metastasis(p<0.01).The expression rates of B-FABP mRNA were not significantly different in associated with gender,age and the size of the tumor(p>0.05).2.35 of 55(63.6%) showed positive B-FABP mRNA signals in blood of patients with RCCC.While only 2 of 28(7.1%) patients in control group showed positive B-FABP mRNA signals.The expression rates of B-FABP mRNA were significantly different between experimental group and control group(p<0.01).The sensitivity and the specificity of the expression of B-FABP mRNA in peripheral blood were 63.6%and 92.9%,respectively.There was no significant correlation with clinical stage,pathological grade and other clinical features.3.The positive expression rates of B-FABP mRNA were 76.4%in tumor tissues and63.6%in blood respectively(p>0.05).Conclusions:1.Tumor tissues of patients with RCCC show significant positive expression of B-FABP mRNA,and the expression is correlated with clinical stage, pathological grade of RCCC.The up-regulated expression of B-FABP mRNA in RCCC suggests that B-FABP mRNA is tightly related with the tumor's occurrence and invasion. Therefore,B-FABP mRNA might be a new index for identifying the prognosis of RCCC.2.The application of RT-PCR-ELISA to detect the expression of B-FABP mRNA in blood shows a high sensitivity and specificity and a minimal invasion.Thus, B-FABP mRNA might be a new index for RCCC diagnosis.3.The expression of B-FABP mRNA in RCCC tissue and in blood shows dependablity. |
