Effect Of Chinese Medicine Psoriasis Prescription No.1 On The JAKs/STATs Signal Pathway In Animal Model Of Psoriasis And Clinical Observation

Objective:To observe the clinical effect of Chinese Medicine Psoriasis Prescription No.1 which is used by Professor Xuan-guowei in the treatment of psoriasis vulgaris usually.To explore the mechanism of Chinese Medicine Psoriasis Prescription No.1 in treating psoriasis vulgaris through animal experiments.Method:1.Animal experiments:(1) First divide 60 Kunming mice and 60 C57 mice into six groups,then give those mice different drugs which including small(the first group),medium(the second group) and large doses of traditional Chinese medicine(the third group),tripterygium glycosides(the fourth group),tripterygium glycosides plus traditional Chinese medicine(the fifth group) and saline(the sixth group) after the animal model was created.Observe the mitotic basal cells in the vaginal epithelium of KunMing mice and the formation of epidermal granular layer in C57 mice tail scales.(2) Take the tail skin of C57 mice and the vaginal epithelium of KunMing mice after they were killed, detect the protein by western blot method(the first antibody adding in the experiment was P-STAT1).(3) Make the tail skin of C57 mice and the vaginal epithelium of KunMing mice into paraffin section,then observe the expression of STAT3,P-STAT3,STAT5 and P-STAT5 in the tissues by immunohistochemistry.2.Clinical research:Divide 78 patients of psoriasis vulgaris into two groups randomly,there were 40 patients in the treatment group and 38 patients in the control group.Give the treatment group Chinese Medicine Psoriasis Prescription No.1 for oral administration and Zilong cream smearing on the rash,the Control group was given tripterygium glycosides tablets to take and Zilong cream.Observe the clinical efficacy after four weeks.Results:1.Animal experiments:(1) Compared with the negative control group, other groups had effected the mitotic in the vaginal epithelium of KunMing mice Obviously(P＜0.05).The third,the fourth and the fifth group had promoted the formation of epidermal granular layer in C57 mice tail scales obviously compared with the negative control group(P＜0.05).(2) The signal size of P-STAT1 on the x-ray film of KunMing mice was decreasing by order as following the fifth group,the fourth group,the sixth group,the third group,the second group and the first group.(3)①In the tail skin of C57 mice,the positive rate of STAT3 in the third and the fifth groups were lower than the other groups; in the vaginal epithelium of KunMing mice,the positive rate of STAT3 in the third and the fourth groups were lower than the other groups.②In the tail skin of C57 mouse,the positive rate of P-STAT3 in the third and the fifth groups were lower than the other groups;there were no significant differences in each group that the expression of P-STAT3 in vaginal epithelium of KunMing mice,③The positive rate of STAT5 in the third,the fourth and the fifth groups were lower than the other groups in the tail skin of C57 mice and the vaginal epithelium of KunMing mice.④In the tail skin of C57 mice,the positive rate of P-STAT5 in the third and the fifth groups were lower than the other groups;there were no significant differences in each group that the expression of P-STAT5 in vaginal epithelium of KunMing mice.2.Clinical research:The total effective rate in treatment group was 75%,and 71.05%in the control group.There were no significant differences between the two groups (P＞0.05),the differences between PASI scores of the two groups were also insignificant after treatment(P＞0.05).Conclusion:1.Chinese Medicine Psoriasis Prescription No.1 can inhibit the epidermal hyperkeratosis and promote the formation of granular layer,so it can improve the pathological state of psoriatic skin.It plays a role in treating psoriasis likely by activating STAT1 or blocking STAT3,P-STAT3,STAT5 and P-STAT5 ways,thus affecting the expression of cytokines.2.Chinese Medicine Psoriasis Prescription No.1 is effective in the treatment of psoriasis and there are no significant side effects.
