| Thyroid cancer is the most common tumors of the endocrine system. Differentiated thyroid carcinoma in papillary carcinoma and follicular carcinoma is a low-grade cancer, good prognosis, if treated timely and reasonable, and is expected to cure the vast majority of patients. But not differentiation of cancer, squamous cell carcinoma, medullary carcinoma is not, of its students belong to a high degree of dioxin cancer, the rapid development and poor prognosis. In addition to surgery, iodine 131 treatment is still a lack of other effective treatment. In recent years, the study found that the incidence of tumor development and is closely related to apoptosis, inhibition of apoptosis regulation of apoptosis genes and pathways, to achieve the purpose of tumor therapy. Stat3 and Survivin as apoptosis-related gene, involved in the regulation of apoptosis.Stat3 was found as a cancer gene in 1990, for an important member of STAT family, Stat3 signal transduction pathway and cell proliferation, differentiation and apoptosis was closely to the continued activation of the pathway can lead to abnormal cell proliferation and malignant transformation. It was well knowed that leukemia, mutiple myeloma, head and neck squamous cell carcinoma, multiple melanoma, breast cancer, prostate cancer and lung cancer were high expression of Stat3. Survivin was identification as new inhibitor of apoptosis gene in 1997, and as one family members of IAP, with inhibition of apoptosis and regulation of cell proliferation in a doubel role. Survivin was not only involved in apoptosis inhibition, cell cycle regulation, tumor angiogenesis, but also widely expressed in many tumor cells.Objective: To detect the expression of Stat3 and Survivin in thyroid carcinoma. And to analyze its relationship with clinicopathologic characterstic.Methods: Immunohistochemical SP method was used to detect Stat3 and Survivin expression. We used X2-test to analyze the result. Select the First Hospital of Jilin University of thyroid surgery in 2007-2008 clinical andPathological data of 42 cases of complete radical resection of thyroid cancer tissue samples and 25 benign thyriod specimens,the patient did not undergo any pre-operative anti-tumor treatment. Thyroid cancer 18 cases were male, 24 cases of women. The age of the cancer between 15 to 69 years old,the average age was 45.6. The age of benign thyriod between 16 to 72 years old,the average age was 30.5. Papillary carcinoma 28 cases, follicular carcinoma 10 cases, medullary carcinoma 2 cases. 28 cases with lymph node metastasis, 14 without.Result: (1) In the 42 cases of thyroid carcinoma, the positive rate of expression of Stat3 and cancer adjacent tissues were 78.6% (33/42), 66.7% (28/42). In the 25 cases of thyroid benign lesion ,the positive rate of expression of Stat3 was 32.0% (8/25). The differences between benign lesion and malignant or benign lesion and cancer adjacent tissues was significant (P<0.05). In the 42 cases of thyroid carcinoma, the positive rate of expression of Survivin and cancer adjacent tissues were 73.8% (31/42), 59.5% (25/42) .In the 25 cases of thyroid benign lesion, the positive rate of expression of Survivin was 24.0% (6/25). The differences between benign lesion and malignant or benign lesion and cancer adjacent tissues was significant (P<0.05). (2) The expression of Stat3 and Survivin was no significant relationship between age, gent. (3) The expression of Stat3 and Survivin was significant relationship on lymph node metastasis. (4) The expression of Stat3 was correlated positively with the expression of Survivin (r=0.384, p<0.05)Conclusions: (1) Stat3 and Survivin in thyroid carcinoma was significantly increase, can be used as diagnostic indicator of benign and malignant, may become the iRNA targeting gene therapy. (2) The expression of Stat3 and Survivin had no difference between age, gent. But was different with lymph node metastasis. (3) Stat3 gene expression was positive correlated with Survivin gene expression. |
