The Experimental Study Of Different Volume Hemofiltration On Acute Renal Failure Animal Model

Objective:To investigate the curative effect and tolerance of different volume hemofiltration on acute renal failure animal model with contrast,and to probe into the safety,efficacy and stability of High Volume Hemofiltration(HVHF) with JH-3038 Blood Purification System in neonates.Methods:24 rabbits(2.20-2.60kg) with acute renal failure were randomly divided into low volume group,middle volume group and high volume group.All animals accepted continuous veno-venous hemofiltration(CVVH) with JH-3038 Blood Purification System,10 hrs in therapy time and 8 ml/min·kg in blood flow rates.The hemodynamic condition and the blood biochemical indicator were monitored during the procedure.The urea clearance under different volume hemofiltration were measured and calculated.The safety and stability of the performance with JH-3038 Blood Purification System were monitored during the procedure.Results:HR,BR and BP of the rabbits in three groups varied with the time(P＜0.05),but the tendency of the change in HR and BR between three groups was no significant difference(P＞0.05).In BP,high volume group was not more undulatory than the other two groups(P＜0.05). Electrolytes were not obviously change pre and post the therapy. Compared with post-treatment,BUN and SCr were conspicuous degraded,there was statistical significant in three groups(P＜0.05).High volume group surpassed low and middle volume group.The rates of urea clearance were 1.32±0.27ml/min,2.61±0.30ml/min and 5.54±0.46mi/min respectively under different volume hemofiltration.The deviation rates of treatment dose were 1.91%,2.02%and 2.39%respectively in three groups.The deviation rate of ulterfiltration volume was 2.48%.During the therapy,JH-3038 Blood Purification System could operate normally and not occur any breakdown.Conclusions:The ARF animal model could be successfully reproduced by the mechanism of the drug poisoning.Under monitoring intently,selecting the equipment justifiably,adjusting the prescription of the treatment dose and setting up the appropriate operation,animals(2.0-3.0kg) could keep the balance of the signs of life during HVHF.High volume hemofiltration could surpass low and middle hemofiltration for stabilizing the hemodynamic condition,purging the solute and electrolyte disturbances and adjusting the acid-base imbalance. Basically,JH-3038 Blood Purification System could conform to the condition of the clinical operation,and be many excellences for the safety,stability,efficacy of HVHF in neonates.
