| Objective:Study the possible relationship between the occurrence of human renal cell carcinoma and the Notch signaling pathway.To explore the development of the molecular mechanism of human renal cell carcinoma for the clinical treatment and provide the theoretical basis.Method:1.Surgical access to renal cell carcinoma specimens,specimens preserved in liquid nitrogen for the tissue.2.Using RT-PCR detection the expression lever of Notch receptor, Notch-1,Notch-2,Notch-3 and Notch-4 as well as the Notch ligand Jagged-1,Jagged-2,Delta-1,Delta-3 and of Delta-4 mRNA.3.On the results of RT-PCR semi-quantitative analysis.Results:1.RT-PCR results showed that the express of receptor of Notch signaling Notch-2,Notch-3 in human cancer patients with renal cell carcinoma tissues higher than in adjacent normal tissue adjacent to cancer, the difference has statistical significance. 2.RT-PCR results showed that the express of Ligand of Notch signaling Notch-2,Notch-3 in human cancer patients with renal cell carcinoma tissues higher than in adjacent normal tissue adjacent to cancer, the difference has statistical significance.3.RT-PCR results showed that the express of another receptor of Notch signaling Notch-2,Notch-3 in human cancer patients with renal cell carcinoma tissues was uncertainty.Discussion:Renal cell carcinoma(renal cell carcinoma,RCC) are the most common adult kidney cancer,primary renal tumor accounting for 80% -90%,and its incidence is still increasing year by year.The natural process of the disease extremely difficult to predict,the overall 5-year survival rate was 40-50%.Therefore,to explore new tumor markers contribute to a deeper understanding of the disease RCC development process and reveals the molecular biology related information to help for RCC diagnosis,treatment and prognosis.The results show that,Notch signaling pathway and the occurrence of renal cell carcinoma must have relevance,Notch signaling pathway and receptor family ligand and the occurrence of renal cell carcinoma has to a certain degree of relevance, one of the Notch receptor signaling pathway Medium,Notch-2 and Notch-3 receptor and the occurrence of renal cell carcinoma showed a positive correlation,Notch signaling pathway ligand Jagged-1 and the occurrence of tumor showed a positive correlation.Prompt the occurrence of renal cell carcinoma and the Notch signaling pathway must exist a possible link.Conclusion:1.There are a certain correlation between Notchsignaling pathway molecules and the occurrence of renal cell carcinoma.2.It showed a certain degree of correlation between Notch receptor signaling pathway of Notch-2,Notch-3 and the occurrence of renal cell carcinoma.3.It showed a certain degree of correlation between Notch ligand signaling pathway of Jagged-1 and the occurrence of renal cell carcinoma4.There are not obvious correlation between Notch signaling pathway ligand Delta-1 and the occurrence of renal cell carcinoma... |
