Much attention has been focused on natural polysaccharides because of its important status in life. Series of Glycyrrhiza polysaccharides (GPS) were prepared from Glycyrrhiza inflate, and their Bio-activities on immuno-regulation, tumor-inhibition and anti-aging function were studied. Futhermore the structure of purified GPS1 was analyzed.The above studies could provide references for application and exploration of Glycyrrhiza inflate resources and protential medical value of Glycyrrhiza polysaccharide.The extraction and separation technology were determined through optimizating optimal processing parameters, and the content of Glycyrrhiza polysaccharide was analyzed. The optimal extracting technology was extracting 2 times with 15 doubles of medicine in 2 h per time on 80℃. The optimal sedimentation technique of Glycyrrhiza polysaccharide was using ethanol as precipitator, concentrating the extract liquid to 2/5 times of its original volume, and adding 4 times ethanol of concentrated liquid. The optimal technique of removing protein from Glycyrrhiza polysaccharide was using the method of trichloroacetic acid to remove protein twice. The optimal decolorization was removing coloring matters by 0.4% absorbent charcoal. The optimal drying technology was low temperature freeze-drying. The content of Glycyrrhiza polysaccharide was determined by phenol-sulfuric acid method. This method was characterized by high degree of accuracy, low relative standard deviation, excellent reproducibility and high recovery of the added sample. The content of polysaccharide in Glycyrrhiza inflate was 1.43%.Using DEAF-cellulose to analyze the components of Glycyrrhiza polysaccharide and four kinds of polysaccharide components were measured. GPS1 was purified trough dialysising and chromatography on Sephadex G-100. The result showed its relative molecular was 6.6KD. From the result of FT-IR, GPS1 may beα-D-pyrane polysaccharide.The activities of Glycyrrhiza polysaccharide were tested by several types of models to study the effects of polysaccharide from Glycyrrhiza on specific, humoral, cellular immunity, immune Organs, anti-tumor and anti-senility in mice. The results indicated that Glycyrrhiza polysaccharide could promote serum hemolysin IgM and IgG production, increase antibody-producting cell level, enhance the macrophage engulfing carbon granula ability, improve the thymus and spleen index in normal mice, add the contents of IL-2 and TNF-α,decrease the content of NO in serum of immunosuppressive mice , inhibit the growth of sarcoma 180 in tumor-bearing mice , and improve the content of SOD,decrease the content of MDA in serum of aging mice.There were significant differences between the control groups and treatment groups. Therefore, Glycyrrhiza polysaccharide has great enhancing immune, anti-tumor and anti-senility function. All these results provided afundamental for further development of clinical application.
