Part IThe Expression and Activation of STAT3 during the Peri-implantation Period of Mouse EmbryoObjective: To investigate the relationship between the expression and activation of STAT3 and mouse embryo implantation.Methods: Immunohistochemistry was used to detect the expression and activation of STAT3 in the uterine endometrium during mouse embryo implantation. Result: Strong level of STAT3 and P-STAT3(the activated form of STAT3) was detected in the luminal epithelium at the midnight of day 4 of pregnancy. The value of the gray scale of group D4 was compared with the value of the gray scale of the other groups. There is significant differences between D4 and the other groups (P<0.05).Conclusion: The time of expression and activation of STAT3 is coincident with the time of mouse embryo attachment and implantation. These data suggests that STAT3 and P-STAT3 play important roles in mouse embryo attachment and implantation.Part IIHormonal regulation of STAT3 expression and activation in mouse uterine endometriumObjective: To investigate the hormonal regulation of STAT3 expression and activation in mouse uterine endometrium.Methods: The ovariectomized mice were divided into four groups. The control group was treated with oil, the E2 group was treated with Estradiol Benzoate, the P group was treated with progesterone, the E2+ P group was treated with estrogen and progesterone. The RT-PCR was used to detect the expression of STAT3 mRNA in the uterus of the four groups. Immunohistochemistry was used to detect the expression of STAT3 and P-STAT3 in the uterine endometrium of the four groups.Result: STAT3 mRNA expression was strongly detected in the uterus of the E2 group and the E2+ P group, while there was no STAT3 mRNA expression in the uterus of the control group and the P group. High level of STAT3 was detected in the uterine endometrium of the E2 group and the E2+P group, while STAT3 was not detected in the uterine endometrium of the control group and the P group. P-STAT3 was not detected in the uterine endometrium of the control group, and it was detected in the uterine endometrium of the other three groups in high level.Conclusion: The expression of STAT3 mRNA and protein in the uterine endometrium were stimulated by estrogen. Progesterone has no use in this, but it can stimulate the phosphorylation of STAT3. Estrogen can stimulate the phosphorylation of STAT3 too.
