Objective:Observing human mononulear apoptosis and assessing TNF-αsevetion by time,after cells cultured with metal particulate and metal ion.Discussing mechanison of osteolysis induced by debris in artificial joint aseptic loosening.Method:Sterilized metal ion and particulates melt in injecting water;confiring asepsis before experiment,using lymphofolt and percoll gradient centrifugation acquire mononuclear cell from peripheral blood,mononuclear has been cultured in the 24 holes culture plate 4×106 cells per hole.There are 9 groups in the experiment.The production of TNF-αwere assessed by enzyme-linked immuno absorbent,assay at 6h 12h 24h 36h 48h.Detecing the apoptosis by flower cytometer.Result:①C obalt particulate induce mere mononuclear apoptosis and TNF-αsecretion than cobalt ion at 12h;②Chromium and Titanium particulate also induce more mononuclear apoptsis and TNF-αsecrtion than Cobalt ion and Chromium ion;③F errum particulate is not different from Ferrum ion in inducing mononuclear apoptosis and TNF-αsceretion.Conclusion:Chromium and titanium particulate induce more human mononuclear and TNF-αsecretion than metal ion at 24h similarly cobalt particulate induce more human mononuclear and TNF-αsecretion than metal ion at 12h,but Ferrum particulate is not different from Ferrum ion.
