Objective:â‘ Investigate the sleeping effect of electro-nape-acupuncture.â‘¡To object the content and the immunohistochemical location of neurotransmitter in the insomnia rats'brain caused by electro-nape-acu puncture.â‘¢Object effect of electro-nape-acupuncture on learning and memery behavioral in the insomnia rats.Methods: 1.Wistar big mice will be regarded as target, we will choose 40 big mice which are divided into 4 groups at random:blank contral group(10 rats);Model contral group(10 rats);Tianmeng capsule group(10 rats); electro-nape-acupuncture group(10 rats). Duplicate inmosnia model of rats by means of PCPA abdominal injection. The animals in the Tianmeng capsule group and electro-nape-acupuncture group will be treated in 7 days. Then study the results of the treatment by observing the content of 5-HT,5-HIAA,NE in the insomnia rats'brain and the positive of CCK8.2. Wistar big mice will be regarded as target, we will choose 20 big mice which are divided into 4 groups at random(5 rats in every group), Duplicateing inmosnia model of rats by means is same.. The animals in the Tianmeng capsule group and electro-nape-acupuncture group will be treated in 7 days. Water Morris is used, place navigation test and spatial probe test are done in rats. Difference s of Ethology were evaluated.Result: 1 Abdominal injection of PCPA deprives the normal sleep of rats, at the same time, there appears decreas the content of 5-HT and 5-HIAA,there appears increas the content of NE and positive of CCK8 in the brains of insomnia rats is decreases, blank group is compared with model group, the difference is notable(P<0.05),the model is success.2 After treaty of the electro-nape-acupuncture,the decreased contents of 5-HT and 5-HIAA in the insomnia rats has increased to the nomal level ,compared with model group the difference is notable(P<0.05).Tianmeng group is compared with model group,the difference is not notable(P>0.05).Tianmeng group is compared with electro-nape-acupancture,the difference is notable(P<0.05).3. After treaty of the electro-nape-acupuncture,the content of NE is decreses,compare with model group, the difference is notable(P<0.05). Tianmeng group is compared with model group,the difference is notable(P<0.05).Tianmeng group is compared with electro-nape-acupancture,the difference is not notable(P >0.05).4. After treaty of the electro-nape-acupuncture,the positive of CCK8 is decreses,compare with model group,the very difference is notable(P<0. 01).Tianmeng group is compared with model group,the very difference is notable(P<0.01).Tianmeng group is compared with electro-nape-acupancture,the difference is not notable(P>0.05).5.After treaty of the electro-nape-acupuncture,the escape latency decreases.times across platform and percentage of probing time increases, compared with model group the difference is notable(P<0.05). Tianmeng group is compared with model group,the difference is notable(P<0.05).Tianmeng group is compared with electro-nape-acupancture,the difference is not notable(P>0.05).Conclution: The electro-nape-acupuncture can be increased 5 - HT,5-HIAA levels, lower NE played in promoting the role of sleep. The insomnia rats lead to learning and memory impairment. After the electro-nape-acupuncture treatment, resumed after learning and memory power. Experimental proof of the electro-nape-acupuncture,can significantly increase the positive CCK8. mechanism for learning and memory is relevant with the it. This may increase the power of learningand memory-the ability to provide a theoretical basis.
