| Objective:To study the expression of PKR,high risk HPV(HPV16/18)in cervical cancer and pre-malignant lesion,and discuss the significance and the possible mechanism in occurrence and development of cervical cancer and pre-malignant lesion by PKR.Methods:From Feburary,2006 to Feburary,2007,we took patients with cervical cancer(n=37)and patient with cervical intraepithelial neoplasia(CIN,n=56)treated in department of gynecology in Tianjin first central hospital,and compared with healthy donor(n=10).:In the 103 cases,using immuno-histochemical SABC stanining method to detect the expression of PKR and HPV16/18 E6.To analyzing the happening and developing of cervical cancer and pre-malignant lesion and the effect in cervical lesion induced by PKR.Result:â‘ The expression of PKR has no close correlation with the age,pathological types and clinical staging(FIGO 2000)of patients with cervical cancer.The positive rate of PKR expression in cervical cancer tissues was 54.1%(20/37);in CINâ…¢tissues was 25%(5/20);in CINâ… -â…¡tissues was 16.7%(6/36)and in normal cervical tissue was 10.0%(1/10).The positive rate of PKR expression in the cancer group is higher than CIN groups and the normal cervical group(P<0.05).The positive rate of PKR expression in the CINâ…¢group is higher than CINâ… â€”â…¡groups,but there were no significant differences(P>0.05).The positive rate of PKR expression in the CIN group is higher than normal cervical group,but there were no significant differences(P>0.05).â‘¡The expression of HPV16/18 E6 has no close correlation with the age and pathological types of patients with cervical cancer(P>0.05),but has correlation with the clinical staging(FIGO 2000)(P<0.05).The positive rate of E6 expression in cervical cancer tissues was 54.1%(20/37);in CIN tissues was 33.9% (19/56)and in normal cervical tissue was 10.0%(1/10)。The positive rate of HPV expression in the cancer group is higher than CIN group and the normal cervical group(P<0.05).The positive rate of E6 expression in the CIN group is higher than normal cervical group but there were no significant differences(P>0.05).The positive rate of HPV16/18 E6 expression in CINâ… group was 29.4%(5/17);in CINâ…¡tissues was 47.4%(9/19);in CINâ…¢group was 25.0%(5/20).The positive rate of E6 expression in the CINâ…¡group is higher than CINâ… group and CINâ…¢group,but there were no significant differences(P>0.05).â‘¢In the cancer group,the expression of PKR and E6 has inversely correlation(r=-0.554,P<0.05),and PKR protein positive rate in those of E6 protein negtive was signifieanay higher than those of E6 protein positive;In the CINâ…¢group,the expression of PKR and E6 has inversely correlation(r=0.480,P=0.032)and PKR protein positive rate in those of E6 protein positive was signifieanay higher than those of E6 protein negtive;In the CINâ… â€”â…¡group,the expression of PKR and HPV E6 has inversely correlation,but there were no significant differences(r=-0.190,P=0.267).Conclusion:â‘ HPV16/18 E6 was strongly associated with an increased risk of developing cervical cancer.â‘¡The upregulation of PKR has relation to cervical cancer and pre-malignant lesion.â‘¢HPV 16/18 E6 gene has relation to modulate PKR expression in.cervical cancer. |
