Objective: To investigate the changes of the visfatin mRNA expression levels inhuman adipose tissues and the relationship between the visfatin mRNA expression levels and metabolic syndrome(MS).Methods :35 MS patients and 15 aged and sex matched healthy control subjectswere enrolled. MS patients who were diagnosed by 2005 IDF criterion were divided into type 2 diabetes mellitus (15 patients) and non- diabetes mellitus (20 patients) again. At the same conditions,we obtained the omental and abdominal subcutaneous adipose tissues from these patients who were underwent elective abdominal surgery operations.9 Breast and 6 thigh adipose tissues samples were gotten from healthy control subjects without MS and type 2 diabetes mellitus. Expression of visfatin mRNA in adipose tissues were determined by semi-quantitive RT-PCR,while body height,weight,waist circumference,hipcircumference,blood pressure,lipodemia,fasting plasma glucose,fasting insulin etc. were investigated.Results:1 BMI, WHR ,WC, BP,FPG,FINS,TG,hsCRP were significantly higher in MS groups than those in control group.( P<0.01).2 In MS groups with type 2 diabetes mellitus and without type 2 diabetes mellitus ,visfatin mRNA expression in omental adipose tissue were significantly higher than it in the subcutaneous adipose tissue respectively (P<0.01). But no difference exsited in control group.3 The visfatin mRNA expression in omental adipose tissue in MS groups was significantly higher than it in control group (P<0.01).Compared with MS group without type 2 diabetes mellitus , the level of visfatin mRNA expression of MS group with type 2 diabetes mellitus was higher (P<0.01). The visfatin mRNA expressions in subcutaneous adipose tissue were not significantly different among three groups.4 Partial correlation showed that FPG was positively correlated with the visfatin mRNA expression in omental adipose tissue (r=0.3090,P=0.043) .5 The visfatin mRNA expression in omental adipose tissue was higher than it in thethigh adipose tissue (P<0.05) .There were not different among other adipose tissues.Conclusion:1 The level of visfatin mRNA expression in omental adipose tissue is higher than it inthe subcutaneous adipose tissue.2.The level of visfatin mRNA expression in omental adipose tissue in MS groups ishigher than it in control group.3.The expression of visfatin mRNA in omental adipose tissue is correlated with FPG.4.The expression of visfatin mRNA in omental adipose tissue seems to be animportant parameter of metabolic syndrome.5.Visfatin maybe is correlated with MS and glucose metabolism.
