Objective:(1) To investigate the difference of ET-1 in the graft caval effluent after flushing of the liver with the cold preservation time changed.(2) To investigate the relationship of ET-1 in the graft caval effluent after flushing of the liver with the graft quality, and to seek a potential method to evaluate the graft quality before implantation.Methods:The isolated reperfsed rat liver model was used. The changes of endothein-1(ET-1) content in the graft caval effluent and liver, alanine aminotransferase (ALT), aspartate aminotransferase (AST) in reperfused liquid were measured in each group. The morphological changes of sinusoidal endothelia cell under were observed.Result:(1) In the same warm ischemic time, the concentration of ET-1 in the graft caval effluent and liver increased with the prolongation of liver cold preservation, especially in the early cold preservation(P<0.01); while ALT,AST activity had obvious changes(P<0.01) just after long term preservation.(2) Abnormal morphological changes of sinusoidal endothelia cell was more obvious along with the prolongation of cold preservation. Conclusion:(1) The concentration of ET-1 in the graft caval effluent and liver has obvious correlation with the degree of injury of sinusoidal endothelia cell.(2) The concentration of ET-1 in the graft caval effluent can reflect the degree of injury of sinusoidal endothelia cell. There is a significant diagnostic value of Endothelin-1(ET-1) in evaluating the quality of donor liver in rat.(3) The concentration of ALT,AST have little value to evaluate the quality of donor liver early.
