Investigate the mechanism of TCM solution on the rat liver during cold preservation, and screen out the TCM solution that is better than Collins solution. Establish the base of clinical TCM solution preserve liver.2 [Methods]SD rats weighing between 200 and 300 g were in all experiments Rats were allowed free access to food and water before procurement of the liver. The SD rats were divided into four groups in random. Collins solution group served as a control. The other three groups were TCM solution groups. Rat livers were cold preserved for 6, 12, and 18 h in Collins solution (group 1) or TCM solution (group 2, 3, 4] at 0-4℃. Afterwards, hepatocytes ALT, AST and LDH in reperfused solution were monitored and liver pathological changes observed. Trypan blue uptake to observe reperfusion injury to sinusoidal endothelial cells. hepatocyte apqptosis was studied by in situ terminal deoxynucle transferase mediated dUTP-HTC nick end-labeling(TUNEL) technique.3 [Results]3.1.TCM solution groups was much improved as compare with that in Collins solution group, accompanied by more significant decrease in ALT, AST and LDH concentration, suggesting better preservation effect of TCM solution compared to that of Collins solution.3.2.After cold preservation, group 3 had more mile pathological changes compared with that in Collins solution group at the same time. No significant changes were observed in pathological findings between the other two TCM solution groups and Collins solution group.3.3. After cold preservation, the sinusoidal endothelial cells deaths in group 3 was less than that in Collins solution group.
